Zingiberaceae is a perennial,terrestrial herb,widely planted in my country,and has the functions of fragrant stomach,dispelling wind,activating collaterals and removing blood stasis,pain reliever or as condiment.As the main active components of Zingiberaceae,curcuminoids and turmeric oil have strong physiological activities due to the existence of various active groups in their molecules,and have wide application prospects.In this paper,the detection methods of active ingredients curcuminoids and turmeric oil in Zingiberaceae were studied,and the detection standards were established;the extraction process was developed and parameters were optimized,and the purification and physiological activity of curcuminoids were explored.The following results were achieved:1.Different detection methods for effective analysis of curcuminoids were constructed:(1)The standard curve of curcuminoids determined by UV-Vis spectrophotometry was y=0.1177x-0.0103,the correlation R=0.9992,and the analytical range was 0.1~4.0 mg/L;(2)The best conditions for HPLC determination are:acetonitrile-4%phosphoric acid solution was the mobile phase(48:52),chromatographic column Agilent C18,0.8 m L/min flow rate,temperature 35°C,detection wavelength 425nm,and methodological validation,and it was concluded that the method has good precision and high repeatability;(3)Using GC-MS to compare the components of turmeric oil obtained by three different extraction methods,it was found that supercritical CO2 The turmeric oil obtained by the extraction method has the largest content of active ingredients,and the content of terpenoids such asα-zingerene,α-curcumene,and turmeric accounted for 78.48%.2.The extraction process was developed and the parameters were optimized:by exploring the single factor test,PB test,response surface analysis and orthogonal optimization test of two new extraction processes,ultrasonic-assisted alcohol extraction and supercritical CO2 extraction,the results were obtained.the best process parameters.(1)Ultrasonic-assisted alcohol extraction method:extracting agent ethanol 74%,ratio of material and solution 1:15 g/m L,ultrasonic power 210 W,time 40 min,temperature50°C;(2)supercritical CO2 extraction(SFE):pressure 15 Mpa,The temperature is 40°C,the mass ratio of entrainer ethanol to raw material is1:10 g/m L,and the time is 2 h.The yields of curcuminoids and turmeric oil were 7.15%,34.68%and 8.38%,45.12%under the best conditions,respectively,and SFE was found to have higher extraction efficiency and better extraction effect on turmeric oil.3.The separation and purification method of macroporous resin for curcuminoids was established.Through a comprehensive comparison of kinetic curves,static adsorption capacity and desorption rate,and time required to reach equilibrium for six macroporous resins,the best macroporous resin HZ-818 was screened out,and the purification process was optimized.When the volume is 20 m L and the ratio of diameter to length is 1:10,the purification effect of curcuminoids is the best,and the product is analyzed and detected,and the purity of curcuminoids is increased from 14.61%to 54.76%.4.The in vitro antioxidant and antibacterial properties of three curcuminoids were explored,and it was concluded that CUR had the strongest DPPH scavenging ability and antibacterial activity,and had the lowest inhibitory effect on Escherichia coli,Bacillus subtilis and Staphylococcus aureus,the minimum concentrations are 1.5 mg/m L,0.7mg/m L,and 6 mg/m L,respectively. |
