Tumeric has traditionally been used both as medicine as well as foods. Its components such as turmeric oil and curcumin embrace research-confirmed bioactivities such as antioxodation, anticancer, anti-inflammatory, for which the techniques for extracting and purifying turmeric oil and curcumin haved turned into s potlight. A comprehensive research in turmeric oil and curcumin has been performed in order to achieve systematic exploitation of them. Firstly, its about highly-efficient extraction techniques of turmeric, which shall be achieved through identification of chemical components by GC-MS and test of anti-bacteria activity. Secondly, it's about the technique to extract curcumin out of the turmeric extract and also further systematic study on techniques to purifying curcumin through macroporous adsorption resin methods so as to establish technical foundation suitable for industrial mass production of purified curcumin. Research results are as follows:1. The comparative study of the process of turmeric oil extraction by microwave-assisted steam distillation and steam distillation was carried out. The extraction rate of curcuma oil by microwave-assisted steam distillation extracting 2h was high more 27.05% then by steam distillation extracting 4h at the same condition.Microwave-assisted steam distillation is an ideal method method appropriate to extract turmeric oil.Through the single factor experiment, orthogonal experiments, the optimizing process was confirmed:grain size 60 mesh,1:8 solid-liquid ratios, extraction time 2.5h. Under the optimal conditions, extraction rate of curcuma oil was 91.06%.2. The oil was analyzed by GC-MS. A total of 81 constituents were detected. The content of 11 compounds was over 1%, and was constituted 71.32% of the total oil.27 constituents was identified, among whichα-Curcumene(8.53%),α-Zingiberene(5.61%),β-Bisabolene(1.72%),β-Sesquiphellandrene(8.83%), ar-Turmerone (31.21%),α-Turmerone(17.57%),β-Turmerone(11.74%) have higher relative contents. The result showed that, the main components include terpenoid compounds; characteristic components ar-Turmerone,α-Turmerone, β-Turmerone reached 60.52%. The result provide a foundation and ideas for turmeric oil's further purification and application development.3. Using antibacterial circle diameter, minimal inhibitory concentrationas (MIC) index, antibacterial activity of curcuma oil was tested by antibacterial experiment in vitro. The test result shows that curcuma oil individually has significant microbial inhibit bioactivity in three different bacteria: Staphylococcus aureus, Bacillus Subtilis, Escherichia Coli, the MIC was 0.31%, 0.62% and 1.25% respectively; Has no microbial inhibit bioactivity in two different fungi:aspergilluse niger, penicillium sp.4. By the extraction method of organic solvent, the yields of curcumine which use using dregs after microwave-assisted steam distillation extracting, turmeric powders, dregs after steam distillation extracting as raw, were 3.27%, 2.68% and 2.65%, The result show that, if the process is proper, the yield of curcumine can be improved.5. Using dregs after microwave-assisted steam distillation extracting as raw, the optimizing process was confirmed:temperature 60℃, ethanol content 80%, extraction time 2 h,1:10 solid-liquid ratios. Under the optimal conditions, yield of curcumin was 3.21%.6. Acrylic aid macroporous adsorption resin was first selected to pure curcumin, the curcumin pure processes was studied by Macroporous Resin Adsorption Process.Resins of adsorption kinetic curve, elution kinetic curve, isothermal adsorption curve of was determined by static experimental, selected a kind of carboxyl resin(Code in the paper was H).Through the single factor experiment, orthogonal experiments, the optimizing process of kinetic adsorption was confirmed:concentration of sample 1.72 mg/mL, flow velocity 1.0 BV/h, ethanol content 60%, volume of sample 8 BV, hydrochloric acid concentration 0 mol/mL, Under the optimal conditions, the adsorption capacity of the H macroporous adsorption resin was 13.84mg/mL resin, adsorption rate was 98.61%.Through the single factor experiment, an orthogonal experiment, the optimizing process of dynamic eluation processing was confirmed:70% ethanol-water as eluent, volume of eluent 17 BV, flow velocity 1.5 BV/h, Under the optimal conditions, the elution rate of curcumin reached 88.36% with a purity 83.12%.Regeneration process of the H macroporous adsorption resin was preliminary studied. By comparing the regeneration effect with 95% ethanol-water,2% HCl- water,5%NaOH,95% ethanol-water was confirmed as optimum regenerated liquid, which optimum volume was 12.0 BV.The experiments of macroporous adsorption resin purification process show that Acrylic aid macroporous adsorption resin purifying curcumin was feasible, and has advantage in adsorption rate, desorption, purity of curcumin. The result laid agood foundation to explore a curcumin pure processes which was fit for industrial application.
