Development Of γ-aminobutyric Acid For The Development Of Complete Nutritional Formula Food For Patients With Refractory Epilepsy

In this study,mice were used as experimental objects to study the alleviating effect of y-aminobutyric acid(GABA),rose concentrated solution and tryptophan(Trp)on epilepsy,and study the alleviating effect of GABA and rose concentrated solution and Trp combination on epilepsy,and the compound of protein,fat,carbohydrate and other nutrients.After optimization,a stable emulsion type of whole nutrition medical food was obtained,and its physiology,toxicity and stability were studied.The changes of Glu:GABA content in the brain of mice with different seizure grades and the indexes of oxidative stress in blood were compared.The higher the seizure grade was,the higher the Glu:GABA content in the brain of mice was,and the more serious the oxidative stress in blood was.When Glu:GABA was higher than 2.77:1,CAT,SOD and GSH-Px activities were lower than 76.65 U/mL,3.28 U/mL and 76.81 U/mL,respectively,and MDA content was higher than 16.68 nmol/mL,epileptic behavior occurred in mice.The oxidative stress indexes(CAT,SOD,GSH-Px and MDA)in blood were determined as the detection indexes of epilepsy in mice.By observing the behavior of mice and measuring the oxidative stress index in blood,the optimal dosage of GABA,Trp and rose concentrate were determined to be 100 mg/kg,160 mg/kg and 1.28 mL/kg,respectively.According to the regulation,the consumption of GABA should not exceed 500 mg/day/person.Screening the combination of GABA-compatible functional substances that can improve epilepsy,The optimal concentrations of GABA+rose concentrate,GABA+Trp,GABA+rose concentrate+Trp were determined to be 10 mg/kg+1.12 mL/kg,10 mg/kg+170 mg/kg,10 mg/kg+1.12 mL/kg+170 mg/kg.According to the efficacy screening of epilepsy animal models,the compatibility of proteins,carbohydrates,fats,vitamins,minerals and other nutrients with GABA and other functional substances for epilepsy patients was determined.With centrifugal stability as the index,through single factor experiment,it was determined that the total nutrition medical food for epilepsy relief contained 4.75%protein,7.75%fat,3%carbohydrate and proper amount of vitamins and minerals.The optimal preparation conditions were as follows:high speed shear at 3500 r/min for 20 min,homogenization at 100 MPa for 4 times,sterilization at 121℃ for 10 min.Four kinds of total nutritional medical food with epileptic relief in mice were obtained:containing Trp(medical food 1),containing GABA+Trp(medical food 2),containing GABA+rose concentrated solution(medical food 3),and containing GABA+rose concentrated solution+Trp(medical food 4).The stability,toxicology and hygiene of four kinds of medical food were studied.The acute oral toxicity test showed that the products were non-toxic.After four times of accelerated stability tests in three months,according to the principles of national health food registration,the shelf life of the product can be considered to be stable within 24 months.Epilepsy relieving efficacy of the four products was tested.Through the behavior of mice and oxidative stress indexes in blood,it was confirmed that the four products had no significant effect on the body weight of mice,the incubation period of epileptic seizure of mice was prolonged for 1 min,and the activities of SOD,CAT and GSH-Px in the blood of mice were increased,while the content of MDA was decreased.This study combined with new nutrition knowledge and the advanced food processing technology,the preparation of 4 full nutrition medicinal food,has a good effect,the ease of epilepsy can also meet the needs of the normal nutrients at the same time,advanced technical theory,obvious effect,with independent intellectual property rights,full nutrition medicinal food can not only supply nutrients required for patients daily.Can also achieve the effect of relieving epilepsy.Compared with surgical treatment,it is safer,and can be combined with drug treatment to achieve the effect of relief.Antiepileptic special type of whole nutrition medical food has broad market prospect,has scientific research and economic significance,and has more health and social benefits.
