Study On The Development Of Nutritionally Complete Food Containing Ginseng SDF

In this study,Ginseng water-soluble dietary fiber,Oligofructose,Whey protein,Soybean protein,Flaxseed oil,Medium-chain triglyceride powder,Vitamin complex and Minerals were used as experimental materials to design three kinds of nutritionally complete foods,including basic formula,Fructo-oligosaccharide formula and Ginseng water-soluble dietary fiber formula(Ginseng SDF formula,the same below).The nutritional improvement effects of three kinds of nutritionally complete foods were compared by nutritional intake deficiency rat model,and the effects on intestinal flora and short-chain fatty acids were evaluated by 16 sr RNA High-throughput sequencing and gas chromatography.At the same time,the stability analysis,safety evaluation and product evaluation of ginseng SDF fully nutritious formula food were carried out.The purpose of this study is to provide a theoretical basis for the research and development of ginseng SDF nutritionally complete foods.1.Referring to the General Rules for Special Medical Use Formulas(referred to as the General Rules for Special Medical Use,the same below)and the Dietary Nutrient Reference Intakes for Chinese Residents as the basis for formula design and preparation,three kinds of whole nutritional formulas are designed.The nutritional improvement effects of three kinds of nutritionally complete foods were studied by animal experiments.Rats in the normal group(protein 12.5%,fat 4%,ash 3.89%),Other groups(protein 10%,fat 3%,ash 3.11%,fat 3%.The model was made for 5 weeks,and body weight and serum albumin were used to judge whether the model was successful or not.After successful modeling,three kinds of nutritionally complete foods were given for 30 days.Compared with the normal group,the body weight of rats in the model group decreased significantly(P < 0.01).,and the liver index,thymus index and spleen index decreased significantly(P < 0.01).The total number of red blood cells,hemoglobin,white blood cells,lymphocytes and neutrophils decreased but there was no statistical difference.Serum total protein and albumin decreased significantly(P < 0.05,P < 0.01),prealbumin decreased no significant difference,and the contents of Ig G and Ig M decreased significantly(P< 0.001).Compared to the model group,the ginseng SDF formulation the body weight gain rate increased significantly(P < 0.01);thymus index and liver index increased significantly(P <0.05);TP,ALB and PA levels increased significantly(P < 0.001);Ig A,Ig G and Ig M levels increased significantly(P < 0.001);The oligofructose formulation group increased the rate of body weight gain(P < 0.05),thymic index and liver index(P < 0.05),and significantly increased the content of ALB and Ig G(P < 0.01,P < 0.01);the basic formulation group significantly increased the rate of body weight gain(P < 0.01),liver index(P < 0.05),ALB,and Ig M(P <0.001,P < 0.05).The results showed that the three kinds of nutritionally complete foods had the effect of improving the nutritional status and regulating the immunity of rats,and the effect of the ginseng SDF formulation was effect.2.The effects of different doses of three kinds of nutritionally complete foods on intestinal flora and fecal metabolites short-chain fatty acids in rats with insufficient nutritional intake were analyzed by 16 Sr RNA High-throughput sequencing and gas chromatography.At the phylum level,compared with the model group,the three kinds of nutritionally complete foods increased the relative abundance of Bacteroidetes and decreased the relative abundance of Firmicutes.At the family level,compared with the model group,the low-dose Fructo-oligosaccharides group and the high-dose ginseng SDF group could reduce the relative abundance of Ruminococcaceae by increasing Lactobacillaceae and S24-7.At the genus level,compared with the model group,each nutritional formula group could regulate the intestinal flora by increasing the relative abundance of Lactobacillus,Clostridiaceae_Clostridium and Akkermansia and reducing the relative abundance of Ruminococcus.Most pronounced in the high dose group of the ginseng SDF formula.The content of short chain fatty acids(SCFAs)in rat metabolites was determined by gas chromatography.The results showed that the content of SCFAs decreased after the model of inadequate nutrient intake was established,with acetic acid,propionic acid and total SCFAs all decreasing significantly(P < 0.001)and isovaleric acid significantly but not significantly.Compared to the model group,total SCFAs in faeces were significantly higher in the oligofructose and ginseng SDF-formulated high-dose groups(P < 0.05,P < 0.01)and acetic acid was significantly higher in the oligofructose and ginseng SDF-formulated high-dose groups(P < 0.05,P < 0.01,P < 0.001),Significant(P < 0.05)levels of propionic acid in all three formulations and significantly increased(P < 0.05)levels of isovaleric acid in the low dose group of the basic formula.The results showed that three kinds of fully nutritious formula foods could maintain intestinal health by increasing the diversity of intestinal flora,the relative abundance of beneficial bacteria and the content of short-chain fatty acids.3.According to the requirements of "Stability Research requirements of Special Medical Formula Food(trial)",the stability of ginseng SDF fully nutritious formula food was evaluated.The results showed that the appearance of ginseng SDF formula food changed in different degrees under three influencing factors,color: high temperature > high humidity > light,caking:high humidity > high temperature > light,and the content of protein and fat did not change significantly under the influence of three conditions.The effect of vitamin E content was high temperature > light > high humidity,but all above the standard line.The acute toxicity test was carried out according to the Technical guidelines for Toxicological examination and Evaluation of Health Food and its Raw Materials(2020 Edition).The results showed that there was no death,abnormal behavior and increased secretion within 14 days,indicating that ginseng SDF formula food belongs to the actual non-toxic level.Through the examination of fungal mycins and microorganisms,the results show that the products meet the requirements of the general rules of special medicine food,all within a reasonable range.This paper studies the ginseng SDF total nutrition formula food developed in the laboratory from the aspects of nutrition improvement and intestinal regulation,and makes a preliminary exploration on the stability,safety and product evaluation of the formula food,which shows that the product meets the requirements of the General rules of Special Medicine.
