Objective: The expression and mechanism of miR-3148 in nasopharyngeal carcinoma(NPC)are currently unclear.This study aims to explore the expression levels and clinical significance of miR-3148 in NPC tissues in Guangxi region,and explore the oncogenic mechanism of action of miR-3148 on NPC through bioinformatics analysis.Methods: Collect the clinicopathological data of 86 NPC tissues and patients from January 2016 to December 2018,and another 25 chronic nasopharyngitis(CNP)tissues from the same period as the control group.Compare the expression differences of miR-3148 between NPC and CNP tissues,and analyze the correlation between miR-3148 expression levels in NPC tissues and clinical pathological characteristics and prognosis of patients.Apply bioinformatics software and dataset GSE53819 to predict and screen differentially expressed target genes,and conduct GO function and KEGG pathway enrichment analysis.At the same time,construct a protein interaction(PPI)network of target genes,identify central target genes,and explore the role of miR-3148 expression imbalance in the occurrence and development of NPC.Results: miR-3148 is highly expressed in NPC tissue(p<0.01),and abnormal upregulation of miR-3148 expression is closely related to distant metastasis of NPC.Patients with high expression may be more susceptible to distant metastasis(p=0.013).The upregulation of miR-3148 abnormal expression can affect the survival rate of NPC patients,and patients with high expression may have lower survival rates(p=0.035).The miR-3148 prediction target gene GO functional analysis found that it was mainly enriched in biological processes such as cell adhesion,positive regulation of gene expression,etc;As well as molecular functions such as transcription factor activity and sequence specific double stranded DNA binding,KEGG pathway analysis mainly involves the PI3K-Akt signaling pathway.PPI network screening for central target genes such as COL1A1,CCND1,FN1,and IGF1.Conclusion: miR-3148 is abnormally upregulated in NPC tissues in Guangxi region.Detecting the expression level of miR-3148 in NPC tissue can help evaluate patient prognosis.MiR-3148 may regulate the PI3K-Akt signaling pathway by targeting COL1A1,CCND1,FN1,and IGF1,playing an important role in the occurrence and development of NPC.
