Diversity Analysis Of Eukaryotic Microalgae In Some Water Area Of Qinghai Province

Microalgae are the main primary producers in water bodies.They are small,fast and widely distributed.In some extreme environmental waters such as high altitude,high salt,low temperature,strong light,high heavy metal content,the microalgae resources are also very rich.Qinghai Province is located in the north of Qinghai-Tibet region.There are many rivers and lakes,including many extreme environmental waters.Conducting a survey of microalgae germplasm resources in the waters of Qinghai Province can provide reliable sampling guidance and data sources for the isolation and research of specific microalgae,and further promote the research on the potential value of algae species,which is of great significance to the development and utilization of local algae resources.This study selected universal eukaryotic primers and used high-throughput technology to analyze the biodiversity of eukaryotic microalgae in 54 different waters areas of Xining City,Haixi Prefecture,Hainan Prefecture,Haibei Prefecture,Huang Guo Shu(Huangnan Prefecture,Yushu Prefecture and Guoluo Prefecture)in Qinghai Province.The sampling scope covers almost the whole of Qinghai.The richness and diversity of sample sites in different regions,the difference of community structure between sample sites,and the composition of microalgae structure and dominant algae genera of microalgae in the sample sites were identified.The main results are as follows:(1)Among the 7 sample sites in Xining City,557 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,belonging to 6 phyla,24 classes,59 orders,95 families,158 genera,and 7 sample points contain 30 different OTUs.The dominant algae are Tetrabaena,Tetraselmis and Desmodesmus.HHSDGY,CDSDGY,QLW and HQSK samples have high abundance,and HHSDGY,CDSDGY,RMGY and HQSK samples have high diversity.The difference of community structure composition between QLW and RMGY and between RMGY and DHMSK is small.The composition of the community structure of HQSK,CDWSCLC and CDSDGY were significantly different from those of the other six sample sites.(2)Among the 8 sample sites in Haixi Prefecture,501 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,belonging to 5 phyla,23 classes,52 orders,89 families,139 genera,8 sample points contain 15 different OTUs.The dominant algae are Tetraselmis,Chlamydomonas and Dunaliella.DLSK,KLKH,CEHYH,XCDMYH samples have high abundance,and CEHYH,DLSK,XSX,CKYH samples have high diversity.The composition of community structure between CKYH and CEHYH and between TSH and XCDMYH is similar.The similarity of community structure composition between KLKH and XSX and XSX and DLSK is small.The community structure composition of GHYH was significantly different from the other seven sample sites.(3)Among the 5 sample sites in Haibei Prefecture,530 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,which belong to 6 phyla,24 classes,61 orders,99 families,164 genera,and 5 sample points contain 42 different OTUs.The dominant algae are Chloromonas,Tetraselmis and Stigeoclonium.The community richness and diversity of DTH samples are the highest.The composition of community structure between GCX and HHXG is similar.The similarity of community structure composition between BYBC and QHH is small.The community structure composition of DTH is quite different from the other four sample sites.(4)Among the 13 sample sites in Hainan,552 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,belonging to 6 phyla,25 classes,60 orders,106 families,170 genera,and 13 sample points contain 9 different OTUs.The dominant algae are Macrochloris,Oophila and Neochlorosarcina.The samples of QZH,WDH,GDHH,STH and AYHC have high abundance,and the samples of WDH,STH,GDHH,LYXSK,AYHC and CSC have high diversity.The composition of community structure among WDH,WQXQL,STH,KH and GDHH is similar.The composition of community structure between LJS and WQXJLS,LYXSK and AYHC,AYHC and GGH,GGH and CSC is similar.The similarity of community structure composition between QZH,LYXSK and AYHC is not obvious.The community structure composition of YDB is quite different from other sample sites in Hainan.(5)Among the 21 sample sites in Huang Guo Shu,773 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,belonging to 6 phyla,25 classes,64 orders,118 families,and 202 genera,and 21 sample points contain 13 different OTUs.The dominant algae are Ulothrix,Mychonastes,and Tetraselmis.ZQH,QSW,ZKH,DQH and JGJD samples have high abundance,and ZQH,QSW,JQH,SKH and QBL samples have high diversity.The composition of community structure between SKH and MKH,ZQH and JQH,ZQH and QSW,ELH and DJCNH,DJCNH and HSXZ,ZLH and NQT,NQT and CLZ,ZLH and XXH,ZLH and LRAC,XXH and LSTYK,CLZ and LBH is relatively similar.The composition of community structure among QSW,QBL,JGJD,GLXGCX,CNKMD sample sites is similar.The community structure composition of NBYZXNH is quite different from other sample sites.(6)Among the 54 locations in Xining City,Haixi Prefecture,Hainan Prefecture,Haibei Prefecture,Huang Guo Shu,1009 OTUs were obtained from taxonomic annotation of eukaryotic microalgae,which belong to 6 phyla,27 classes,73 orders,141 families and 255 genera.The five regions together contain 233 different OTUs,and the abundance of Chlorophyta is more than 90%.
