Freshwater microalgae have different forms,rich species and wide distribution.They are important components of aquatic communities,and also important contributors to the primary productivity of water bodies.They have important ecological functions.Many kinds of freshwater microalgae have high application value and play an important role in the material and energy cycle of freshwater ecosystem.The classification and identification of microalgae is the basis of microalgae research and the beginning of microalgae resource development.Since the 19 th century,the classification methods of microalgae have experienced the development from morphological identification,physiological and biochemical identification to molecular biological identification.Although each method has its limitations,human beings have never stopped the development of microalgae resources in various waters.In this study,we selected specific primers for V4 hypervariable region of ribosomal small subunit 18 srDNA gene of eukaryotes,and used high-throughput sequencing method to carry out a preliminary study on the biological diversity of freshwater eukaryotes in 18 samples from some waters of South Central Hebei Province for the first time.The abundance,structure and dominant species of microalgae were analyzed by using qiime,mothur and usearch software.The main results are as follows:1.In this study,the effective sequence and OTU number of each sample for analysis are as follows: XTZ04 is 53096/320,XAWQ02 is 32005/211,HDZ03 is 36390/290,HDF03 is 38737/258,STP02 is 37425/272,CZRM02 is 31586/278,XABY02 is 64682/286,CZNH02 is 44094/239,HDC03 is 47247/226,XTW03 is 33322/266,shtq02 is 36685/218,XTQ04 is 29450/175,HDL0 4 is 26232/169,XADQ02 is 32713/149,CZSC02 is 27042/140,XTX03 is 36511/159,CZLJ02 is 37629/112,CZNY02 is 29045/66.2.The sparse curve indicates that the sequencing data volume of each sample is sufficient,and the OTU quantity gradually tends to be saturated.3.From the ACE and Chao1 indexes representing the community abundance,it can be seen that the two indexes rank the sample community abundance in the same order.The sample community abundance from large to small is STP02,XABY02,SHT02,XTQ04,XTW03,HDZ03,XTZ04,CZRM02,HDF03,CZNH02,HDC03,HDL04,XADQ02,XAWQ02,XTX03,CZSC02,CZLJ02,CZNY02.4.From the Shannon index representing the species diversity of the community,it can be seen that the sample diversity is from large to small: HDZ03,CZRM02,HDF03,XTZ04,XTW03,XAWQ02,CZNH02,XTX03,CZSC02,STP02,SHT02,XABY02,CZLJ02,HDC03,XTQ04,HDL04,XADQ02,CZNY02.From the Simpson index representing the species diversity of the community,it can be seen that the sample diversity is from large to small: HDZ03,XTW03,XTX03,CZNH02,CZRM02,HDF03,XTZ04,XAWQ02,CZSC02,STP02,CZLJ02,XADQ02,XTQ04,XABY02,HDL04,SHT02,HDC03,CZNY02.5.Community structure and dominant species of microalgae in each sample: HDL04 mainly consisted of 2 phylum,3 class,6 orders,10 families,and 17 genera,among which the dominant species are Acutodesmus?Coelastrum and Neochloris.CZSC02 mainly has 2 phylum,3 classes,3 orders,7 families and 12 genera,among which the dominant species are Coelastrum?Monoraphidium and Golenkinia.XTQ04 mainly consists of 2 phylum,2 classes,4 orders,7 families and 15 genera,among which the dominant species are Acutodesmus?Coelastrum and Desmodesmus.CZLJ02 mainly consists of 2 phylum,3 classes,6 orders,6 families and 11 genera,among which the dominant species are Acutodesmus?Coelastrum and Dictyosphaerium.CZNY02 mainly consists of 3 phylum,4 classes,4 orders,5 families and 7 genera,among which the dominant species are Coelastrum and Ochromonas.XABY02 mainly consists of 1 phylum,2 classes,4 orders,6 families and 12 genera,among which the dominant species are Acutodesmus and Coelastrum.XADQ02 mainly has 1 phylum,1 class,3 orders,5 families and 13 genera,among which the dominant phylum,3 classes,3 orders,3 families and 7 genera,amongwhich the dominant species are Golenkinia and Desmodesmus.HDC03 mainly has 1 phylum,4 classes,4 orders,4 families and 6 genera,among which the dominant species are Ochromonas and Lecane.HDF03 mainly has 3 phylum,4 classes,6 orders,3 families and 10 genera,among which the dominant species are Lecane and Chlamydomonas.XTW03 mainly consists of 1 phylum,3 classes,4 orders,6 families and 10 genera,among which the dominant species are Desmodesmus?Ochromonas and Golenkinia.XTX03 mainly consists of 2 phylum,3 class,4 orders,4 families and 7 genera,among which the dominant species are Chlamydomonas?Characium and Mychonastes.XTZ04 mainly has 5 phylum,5 classes,6 orders,3 families and 6 genera,among which the dominant species are Coelastrum?Choricystis and Mychonastes.CZNH02 mainly consists of 2 phylum,4 classes,4 orders,5 families and 8 genera,among which the dominant species are polymonophyllum,euglenophyllum and rhombocystis.CZRM02 mainly consists of 2 phylum,4 classes,4 orders,4 families and 11 genera,among which the dominant species are Coelastrum and Nitzschia.XAWQ02 mainly consists of 3 phylum,5 class,6 orders,4 families and 8 genera,among which the dominant species are chaetophyta,chlorella and eustachia.
