Research objective:To observe the effect of 6-week HIIT on behavioral and blood flow signaling pathways in APP/PS1 transgenic mice,and to explore the mechanism of HIIT on the pathology of MCI.In order to provide non-drug,safe and effective prevention and treatment strategies for AD,provide experimental basis and theoretical basis for clinical application of HIIT.Research methods: 1.Animal groups: 15 female mice with 12-week-old in each group:(1)C57BL/6J quiet control group(group C);(2)C57BL/6J HIIT group(CT group);(3)APP/PS1 transgenic control group(group M);and(4)APP/PS1 transgenic high-intensity interval exercise group(MT group).2.Exercise program: treadmill exercise was carried out for 6 weeks in CT group and MT group,and the blood lactic acid concentration of mice in CT group and MT group was measured before,during and after training.3.Morris water maze was used for positioning navigation and space exploration test.4.The elevated cross maze test was used to detect the anxiety of mice in each group.5.The expressions of HIF-1?,VEGF,EPO,ET-1,e NOS,GPR68,A? and Phospho-Tau proteins in hippocampus of mice in each group were detected by Western blot.6.Amyloid plaque deposition was observed by amyloid staining(modified Stores Congo red method).7.ALP was used to observe the abundance of blood vessels in the hippocampusResearch results:(1)MT group and CT group increased blood lactate levels during training,Blood lactic acid level drops after training.During exercise,the blood lactate level of MT group was significantly higher than that of CT group.(2)The results of the Morris water maze showed that compared with the incubation period on day 1,there was a very significant decline in the C group on the 3rd and 4th day,the CT group and the MT group began to show a very significant decline on the 4th day,and the M group had no significant difference.The incubation period of group M was higher than that of group C on the 3rd and 4th day,and it was higher than that of MT group on the 4th day.Compared with the percentage of time in the platform quadrant on day 1,there was a significant increase on day 3 in the C and CT groups,a very significant increase on the 4th day,a very significant increase in the MT group on the4 th day,and no significant increase in the M group difference.On the 4th day,the percentage of time in the platform quadrant of the M group was lower than that of the C and MT groups.The ratio of the distance percentage of the platform quadrant of the space exploration group M is lower than that of the C group and the MT group.(3)The results of the elevated plus maze experiment show:Compared with C group and MT group,M group has a decrease in the number of arm opening times,time,and the ratio of total times and total time.Compared with group C,the time to enter the arm opening and the ratio to the total time increased in the CT group.(4)Western Blot:Compared with group C,A?,Phospho-Tau,ET-1,and GPR68 expression in M group increased;VEGF and e NOS decreased.The expressions of A?,Phospho-tau,ET-1,and GPR68 in the MT group were lower than those in the M group;HIF-1?,VEGF,EPO,and e NOS were increased.The expression of HIF-1? and EPO in the CT group was higher than that in the C group.(5)Amyloid staining(congo red staining)found no amyloid plaques in the C and CT groups,amyloid plaques in the MT group in the M group.The MT group has relatively small amyloid plaques than the M group(6)Through ALP staining,it was found that the blood vessel abundance of the M group was lower than that of the C and MT groups.Research conclusion: Depletion of learning and memory in MCI mice,and developed anxiety and depression.A relative decrease in vascular abundance and vascular tone in MCI mice,resulting in energy supply disorders and brain toxin metabolism disorders,and causing amyloid deposition and neuronal entanglement.After the intervention of HIIT,the Cognitive function was improved,Anxiety symptoms improved,the vascular abundance and vascular tension were relatively increased,the toxicity of A? and Tau phosphorylation to the brain was reduced.
