| ObjectiveRetrospective analysis of 601 cases of advanced gastric cancer patients with TCM four diagnosis information,to explore the distribution of TCM syndrome of advanced gastric cancer,and based on network pharmacology to explore the mechanism of TCM treatment of advanced gastric cancer,to provide a new idea for the clinical and scientific research work of TCM treatment of advanced gastric cancer.Methods1.The four diagnosis information of 601 patients with advanced gastric cancer was retrospectively collected,and the investigation results were analyzed by combining factor analysis and cluster analysis,so as to explore the distribution pattern of TCM syndrom of advanced gastric cancer.2.Network pharmacology method is applied to explore the mechanism of action of new good attached party treatment for cancer of the stomach,through the "drug active ingredient-a disease-therapeutic targets," the construction of a network diagram and visualization,target protein interaction network(PPI)build,GO enrichment and KEGG pathway enrichment analysis function,looking for traditional Chinese medicine key targets and pathways for the treatment of gastric cancer.Results1.A total of 10 principal components were obtained through principal component analysis on 29 data of four diagnoses of Chinese medicine.According to the loading factors of each data of four diagnoses of Chinese medicine,the four diagnoses data with factor coefficient>0.4 were selected and allocated to the 10 principal components.Then,based on the results of principal component analysis,cluster analysis was conducted to obtain the distribution proportion of the three types of TCM syndesmes.According to the differentiation of the three types of TCM syndesmes by professional clinicians,the results were followed by the frequency distribution as cold coagulation and blood stasis(59.28%),deficiency of stomach Yin(24.17%)and phlegm heat accumulation(16.55%).2.VEEN software was used to obtain the intersection of drug action targets and disease-related targets,and a total of 87 common targets were obtained.Thirteen active components of Alpinia officinalis,18 active components of Alpinia officinalis,and 3 active components of Pangolin officinalis were screened out.Eighty-seven key target proteins were identified by PPI network.Through GO functional enrichment analysis,92 items were obtained,which mainly involved receptor activity,protein binding,enzyme activity regulation,transcription factor activity,etc.149 enrichment pathways were identified by KEGG pathway enrichment analysis,and the key pathways were PI3K-Akt signaling pathway,p53 signaling pathway,Wnt signaling pathway,etc.Conclusion1.The basic TCM syndromes of advanced gastric cancer fall into three categories,including cold coagulation and blood stasis,deficiency of stomach Yin and phlegm and heat acccumulation.Among them,cold coagulation and blood stasis accounted for the highest proportion.In conclusion,the pathogenesis of advanced gastric cancer is mainly cold coagulation and blood stasis,accompanied by deficiency of gastric Yin and accumulation of phlegm and heat.The treatment should focus on warming the middle and dispersing cold,promoting blood circulation and removing blood stasis,while simultaneously benefiting stomach and nourishing Yin,clearing heat and resolving phlegm.2.New good party attached more importance in the treatment of cancer of the stomach of the composition is probably indicates Jiang Su,quercetin,mignonette,diosgenin,etc,may be mainly through MYC,AKT1,CASP3 and EGFR targets to achieve therapeutic purposes,such as the main channel for PI3K-Akt signaling pathway,p53 signaling pathway and Wnt signaling pathways,may through the protein,activity of transcription factors and enzyme activity,receptor activity adjustment to treat cancer of the stomach,explain the new he attached the treatment of cancer of the stomach is mainly by multiple components,multiple targets,generate synergy effect of treatment. |
