Corni fructus is the dried and mature pulp of Cornus officinalis Sieb.Et Zucc.It has the effects of nourishing liver and kidney,astringent essence and solid removal.It is a traditional medicine commonly used in traditional Chinese medicine to treat diabetes One,Mainly produced in China's Henan,Shaanxi,Zhejiang and other places.Due to the wide range of planting,the great difference of planting base environment and the disunity of harvesting and processing technology,the quality of Corni fructus is uneven in the markets.In view of this,on the basis of the previous textual research,this study screened the evaluation indicators of the appearance characteristics of Corni fructus,combined with market transactions,and formulated the grade standard of Shaanxi Corni fructus products.By introducing the colorimetric principle,the color data of medicinal materials were read by Matlab 2014a software,and the contents of 5 major substances and 7 components in Corni fructus were determined,explore the correlation between the appearance color of Corni fructus and its secondary metabolites.NIR and MIR spectral data of Corni fructus were collected,and the origin identification and content determination of Corni fructus were studied by LR,KNN,BP-ANN and SVM.This study aims to conduct in-depth research on the specifications and standards of Corni fructus decoction pieces,to establish convenient,accurate and stable methods of producing area identification and quality evaluation for quality control and evaluation of Corni fructus.This study provides theoretical basis and technical guidance for production practice.The main results of this paper are as follows:(1)Based on the research of herbal medicine and market research,combined with market transactions,this study suggested that the content of Corni fructus pieces and grain are supplemented for Corni fructus appearance character.This study makes the original region of Corni fructus specifications and grades in Shaanxi,which can be divided into selected goods and currency,and the former are three levels,first,second,third,mainly according to bright red appearance,without impurities,kernels per 150g or higher is preferred.(2)The images of 29 batches Corni fructus were Collect,and the chromaticity value of the images of Corni fructus from different areas was extracted by Matlab 2014a and 5 major substances and 7 components of Corni fructus were determined.Combined with three different chemometric methods such as MLR,BP-ANN and SVM,the correlation between the appearance color color value of Cornus officinalis and its secondary metabolites was analyzed,and the content determination model was established.The results showed that the content of total organic acid Corni fructus was significantly correlated with its color.(3)The NIR and MIR spectra of Corni fructus were collected,and the origin identification model of Corni fructus was established by LR,KNN,BP-ANN and SVM.The results showed that the identification model established by NIR and MIR.spectrum combined with SVM has a high identification rate on the origin of Corni fructus and can be used to identify the origin of Corni fructus.(4)The NIR and MIR spectral were collected,and contents of 7 kinds of ingredients in Corni fructus were detected using HPLC.This study combined with the analysis of LR,BP-ANN and SVM to establish Corni fructus content prediction model based on infrared spectrum.The results showed that the NIR spectroscopy and MIR spectroscopy combined SVM in establishing the content determination of gallic acid,5-HMF,monoside,sweroside,loganin,cornuside in Corni fructus possess high accuracy,which can be used for content determination of active ingredients of Corni fructus.The model based on NIR spectroscopy combining SVM was more accurate in predicting the content of active components in Corni fructus.
