Cornus officinalis is a Macrocarpium plant of the family Cornaceae,which is a deciduous shrub or tree.It is a valuable traditional Chinese medicine.It tastes sour,astringent,lukewarm,returning to liver and kidney essence and has the potent effects in tonifying liver and kidney and strengthen the essence.It can be used in treatment of vertigo and tinnitus,sweating and prostration,internal heat and thirst,lumbar and knee ache,frequency of urinatior and enuresis,impotence and seminal emission,uterine bleeding and so on.At present,the research on the quality standard of Cornus officinalis mainly focuses on the content of the effective chemical components.There is little research on the commodity specification grades of Cornus officinalis and there is no universal reference standard for the classification of commodity specification grades of Cornus officinalis.In this paper,the research object is Cornus officinalis collected from Henan,Shanxi and Zhejiang provinces.The properties,chemicals and biological activities of Cornus officinalis of different grades were evaluated to explore the relationship between commodity grades and properties,chemical components and biological activities in order to draw up a new standard for the commodity specification grade of Cornus officinalis.(1)According to the surface color,texture and impurities,66 batches of Cornus officinalis were divided into four grades.Then,the color and thickness of the peel were measured and recorded.(2)The HPLC was used to determine the loganin and morroniside in Cornus officinalis.Leachate,water extract,50%ethanol and 95%ethanol extract were obtained by cold soaking,heating reflux extraction and ultrasonic extraction and their contents were calculated,respectively.The contents of total pigment,total iridoid glycoside,total phenolic acid and total polysaccharide were determined by UV-Vis spectrophotometry.DPPH and SOD kit methods were used to evaluate the antioxidant activity of Cornus officinalis in vitro.And the anti-inflammatory activity of aqueous extract of Cornus officinalis was evaluated,which on lipopolysaccharide(LPS)-induced NO production in murine monocyte macrophage(RAW 264.7)was investigated by LPS-induced RAW 264.7 model.(3)The SPSS statistical analysis was used to analyze the datas.Exploratory analysis tests outliers in 66 sets of data and the normality of the data.Factor analysis integrated 11 chemical measurement indicators into 3 common factors,which were named as indicator component factor,extraction factor and category factor respectively.8 related indicators were selected after multiple optimization and combination,and clustering analysis was performed on 62 Cornus officinalis samples to obtain the re-classification results of 62 samples by average connection dendrogram,and the classification results of all the reference multi-variable combinations were obtained.Finally,a new grading standard of Cornus officinalis was developed.
