| Fructus corni,a herbal medicine,abundant in China,has various biological activities.In the thesis we investigated the extraction,separation,purification and physicochemical properties of the polysaccharides from fructus corni,we also studied the antioxidant activities and Immunity activities of the polysaccharides.We studied two extraction methods,ultrasonic technology and boiling water technology,in which we extracted polysaccharides from fructus corni.Several factors that affecting the extraction process were studied through one-factor experiment and orthogonal experiment.The optimal technological conditions of water extraction from fructus corni were decided:extraction temperature of 80℃,ratio of solid to liquid 1:60,extraction duration of 5h,extraction times of 2. The optimal technological conditions of ultrasonic extraction were determined:sound power 400w,extracting time 40minutes,ratio of solid to liquid 1:60.From the results:we found that the method of ultrasonic extraction is more efficient,more sufficient extracting,less energy sources consumingTwo purified polysaccharides S2(extracted by ultrasonic) and F4(extracted by water) were prepared with process as the following:water(ultrasonic) extracting,ethanol precipitation,clearing protein by Sevag way,DEAE-cellulose column and Sephadex G-200 column chromatography.They have the same properties of the polysaccharides.The antioxidation activities of the polysaccharides were studied by scavenging superoxide anion free radicals,hydroxyl free radicals and DPPH free radicals.The results showed that the polysaccharides had better effects of elimination of·OH,O2·- and DPPH.In immunological test,we find S2 in vitro showed directly promote the survival of spleen cell and can synergize the proliferation by PHA and ConA.But S2 in vitro can not enhance macrophage phagocytosis. |
