Association Of Vascular Endothelial Growth Factor Receptor 2 And Gelatinase Genetic Polymorphisms With The Susceptibility Of Recurrent Spontaneous Abortion: A Systematic Review And Meta-analysis

Objective The aim of this study is to evaluate the association between vascular endothelial growth factor receptor 2(VEGFR2;kinase insert domain receptor,KDR)rs6838752,rs2071559,rs2305948 and rs1870377 single-nucleotide polymorphisms and the susceptibility of recurrent spontaneous abortion(RSA)by systematic review and meta-analysis comprehensively.Methods Databases including Pub Med,EMbase,Web of Science,Scopus,Cochrane Library,CNKI,CBM,VIP and Wan Fang were searched from inception to October 2020,and the case-control studies about the relationship between KDR genetic polymorphism and RSA were collected.Two researchers independently screened the literatures,and evaluated the quality of the included literatures according to the NOS scale.Then,heterogeneity analysis and data extraction were performed,and the Stata software was applied for meta-analysis on the studies that met the inclusion criteria in the form of binary data.The observation indicators are the combined odds ratio(OR)and its 95%confidence interval(95%CI).P<0.05 was considered statistically significant.This study has not been registered on any platform.Results A total of 8 case-control studies were included in the systematic review,involving 6single-nucleotide polymorphisms of rs6838752(C/T),rs2071559(T/C),rs2071559(A/G),rs2305948(C/T),rs2305948(G/A)and rs1870377(A/T).There was a study respectively showing that rs6838752(C/T),rs2071559(A/G),rs2071559(T/C)and rs2305948(G/A)polymorphisms were related to RSA susceptibility.There was a study respectively showing that rs6838752(C/T)and rs2071559(T/C)polymorphisms had nothing to do with RSA susceptibility.2 studies respectively showed no association between rs2305948(C/T)and rs2305948(G/A)polymorphisms and RSA susceptibility.2 studies showed that rs1870377(A/T)polymorphism was related to RSA susceptibility and 5 studies showed that rs1870377 polymorphism had nothing to do with RSA susceptibility.A total of 5 case-control studies were included for meta-analysis,including 832 cases in the case group and 811 cases in the control group,all of which were Asians.The result showed that for the KDR rs1870377(A/T)polymorphism in Asian populations,only recessive gene model was statistically significant between the case group and the control group[TT vs.AT+AA:OR=1.34,95%CI(1.07,1.68),P=0.01,I~2=0%].Conclusions KDR rs1870377(A/T)polymorphism was associated with the susceptibility of RSA in Asian populations.Objective The aim of this study is to evaluate the association between matrix metalloproteinase 2(MMP-2)rs243865(C/T),rs2285053(C/T)and matrix metalloproteinase 9(MMP-9)rs3918242(C/T)single-nucleotide polymorphisms and the susceptibility of recurrent spontaneous abortion(RSA)by meta-analysis comprehensively.Methods Databases including Pub Med,EMbase,Web of Science,Scopus,Cochrane Library,CNKI,CBM,VIP and Wan Fang were searched from inception to August 2020,and the casecontrol studies about the relationship between gelatinase genetic polymorphisms and RSA were collected.Two researchers independently screened the literatures,and evaluated the quality of the included literatures according to the NOS scale.Then,heterogeneity analysis and data extraction were performed,and the Stata software was applied for meta-analysis in the form of binary data.The observation indicators are the combined odds ratio(OR)and its 95% confidence interval(95%CI).P<0.05 was considered statistically significant.This study has not been registered on any platform.Results A total of 6 case-control studies were included in meta-analysis.The result showed that for the polymorphism of MMP-2 rs243865(C/T),there was no significant difference between the RSA group and the control group.For the MMP-2 rs2285053(C/T)polymorphism,only the allele,heterozygous and dominant gene models were statistically different [T vs.C: OR=1.50,95%CI(1.08,2.10),P=0.02,I2=31%;CT vs.CC: OR=1.72,95%CI(1.07,2.77),P=0.02,I2=41%;CT+TT vs.CC: OR=1.73,95%CI(1.10,2.70),P=0.02,I2=41%].For the MMP-9 rs3918242(C/T)genetic polymorphism in overall population,there were no statistically significant differences in allele,homozygous,heterozygous,dominant and recessive gene models.For the MMP-9 rs3918242(C/T)gene polymorphism in Asian population,the allele,heterozygous and dominant gene models had statistically significant differences [T vs.C: OR=1.38,95%CI(1.12,1.69),P=0.002,I2=0%;CT vs.CC: OR=1.45,95%CI(1.12,1.87),P=0.004,I2=0%;CT+TT vs.CC: OR=1.48,95%CI(1.16,1.89),P=0.002,I2=0%].Conclusions MMP-2 rs2285053(C/T)polymorphism was related to the susceptibility of RSA.The polymorphism of MMP-9 rs3918242(C/T)was not associated with the susceptibility of RSA in the overall population,but it was related to the susceptibility of RSA in Asian population.
