Assocition Between Vascular Endothelial Growth Factor Gene Polymorphisms And Risk Of Recurrent Spontaneous Abortion:a Meta-analysis

Objective:To evaluate the association between vascular endothelial growth factor (VEGF) gene polymorphisms and risk of recurrent spontaneous abortion (RSA).Methods:Literatures published on PubMed, EMbase, Cochrane Library, CKNI, WANFANG DATA and SinoMed were searched based on the established strategy.Quality of included studies was assessed,the analysis of heterogeinety between the included studies was performed.ORs and their 95%CI were used to assess the strength of association between VEGFgene polymorphisms and risk of RSA. Stata 12.0 was used to analyze the data.Results:A total of 13 case-control studies were eligible for the meta-analysis. Among these studies,nine studies with 1608 cases and 1804 controls studed on rs 1570360, nine studies with 1520 cases and 1705 controls studed on rs3025039, seven studies with 1282 cases and 1475 controls studed on rs2010963, eight studies with 1431 cases and 1514controls studed on rs699947.Our meta-analysis suggests insignificant association between rs699947 polymorphism and risk of RSA,with (OR=1.01,95%CI: 0.90-1.12),(OR=1.05,95% CI:0.90-1.22), (OR=1.02,95% CI:0.82-1.27) under allele, dominant and recessive genetic models,respectively.But women with allele T or genotype TT in the rs3025039 polymorphism have higher risk of RSA,with(OR=1.38,95% CI:1.20-1.59),(OR=1.42,95% CI:1.21-1.67), (OR=1.57,95% CI:1.01-2.43) under allele, dominant and recessive genetic models,respectively. Women with allele C or CC genotype in the rs2010963 polymorphism have higher risk of RSA, with(OR=1.14,95% CI:1.02-1.27), (OR=1.30,95% CI:1.03-1.63) under allele, codominant(CC vs. GG) genetic model, respectively. Women of non-Asian regions with AA genotype in the rs 1570360 polymorphism have higher risk of RSA under recessive and codominant (AA vs. GG、AA vs. GA) genetic models(OR=2.26,95% CI: 1.50-3.41; OR=2.38,95% CI:1.13-5.02, and OR=2.15,95% CI: 1.38-3.36,respectively). but our meta-analysis results show an insignificant association between rs1570360 polymorphism and RSA of Asian regions under all genetic models.Conclusions: - 2578C/A (rs699947) polymorphism is not significantly associated with the risk of RSA, whereas +936C/T (rs3025039) and 634G/C(rs2010963)polymorphisms show significant associations with RSA. Women with allele T or genotype TT in the +936C/T (rs3025039), allele C or genotype CC in -634G/C(rs2010963) polymorphism have higher risk of RSA. Women of non-Asian regions with AA genotype in the -1154G/A(rs1570360) polymorphism have higher risk of RSA,but not with that of Asian regions.
