Objective:This study aims to develop a convenient air disinfectant of high efficiency and low toxicity through verifying the effectiveness and safety of TCM air disinfectant based on Qingwen Bizhuo Recipe,so as to provide a new conventional disinfectant for epidemic prevention and control.Methods:The effectiveness of air disinfectant was verified by laboratory disinfection test and clinical air disinfection experiment.The safety of air disinfectant was verified by animal safety experiment.The subjective adverse reactions caused by air disinfectant were preliminarily studied by questionnaire survey.1.Laboratory disinfection test:The inactivation capacity of the TCM air disinfectant against staphylococcus aureus,escherichia coli and candida albicans was tested by a suspension quantitative test,and a neutralizer identification test was used to verify the effectiveness of TSB containing 2%of lecithin and 2%of Tween 80 in neutralizing the residues of TCM air disinfectant.2.Animal safety test:The inhalation toxicity of the TCM air disinfectant was evaluated by an acute inhalation toxicity test in mice,and the irritation effects to the skin and eye were determined respectively by the skin irritation test and eye irritation test respectively in rabbits.3.Clinical air disinfection experiment:The six-stage sieve impingement air sampling method and settling-plate method were used for sampling and cultivation,so as to determine the colony numbers in air before and after use of the TCM air disinfectant,calculate the killing rate,determine the environmental standard grade after disinfection,and evaluate relevant air disinfection effect in the environment.At the same time,a comparison of disinfectant effect was conducted between the TCM air disinfectant and peracetic acid.4.Adverse reactions:Considering that the disinfectant may cause discomfort of eye,skin and respiratory system,digestive system,and etc.,a preliminary study on possible subjective adverse reactions caused by the TCM air disinfectant was carried out through a questionnaire survey,and at the same time,a comparison between the TCM air disinfectant and peracetic acid in severity of adverse reactions was conducted.Results:1.Laboratory disinfection test:In the suspension quantitative test,the killing log values of the TCM air disinfectant against staphylococcus aureus,escherichia coli and candida albicans were greater than 5 for all groups,indicating that the TCM air disinfectant had an obvious bactericidal effect.Furthermore,the TSB containing 2%of lecithin and 2%of Tween 80 neutralized the residues of TCM air disinfectant effectively.2.Animal safety test:A maximum inhalation toxicity test in mice showed that the TCM air disinfectant had no inhalation toxicity,and the skin irritation test and eye irritation test in rabbits showed that the TCM air disinfectant was non-irritating to the skin and eyes.3.Clinical air disinfection experiment:There was a statistically significant difference in the total number of colonies in the air before and after use of TCM air disinfectant(p<0.05,n=20),and the killing rate was more than 90%,which indicated that the air disinfection capacity of the TCM air disinfectant based on Qingwen Bizhuo Recipe was acceptable.The number of colonies based on settling-plate method for sampling was less than 4.0 CFU(5min),which met the requirements for Class Ⅳ environment.When there was no statistically significant difference between the TCM air disinfectant and peracetic acid in the number of colonies before disinfection(P>0.05,n=60),there was 30min after disinfection(P=0.008<0.05,n=60),indicating that the TCM air disinfectant based on Qingwen Bizhuo Recipe was inferior to peracetic acid in disinfection capacity.4.Adverse reactions:In the questionnaire survey based on 30 person-times each group,after use of TCM air disinfectant,no subjective discomfort reaction was reported,and after use of peracetic acid,there were 21 person-times of systemic irritation.Therefore,the TCM air disinfectant was preliminarily evaluated as superior to peracetic acid in subjective adverse reactions.Conclusion:1.The TCM air disinfectant based on Qingwen Bizhuo Recipe may kill staphylococcus aureus,escherichia coli and candida albicans effectively.2.The TCM air disinfectant based on Qingwen Bizhuo Recipe has no inhalation toxicity to mice and no irritation to the skin and eyes of rabbits.3.The TCM air disinfectant based on Qingwen Bizhuo Recipe has met the "Technical Standard for Disinfection" for field disinfection capacity,and the emergency rooms may meet the requirements of Class Ⅳ environment after disinfected by it,but for the disinfection effect,it is inferior to peracetic acid.4.The TCM air disinfectant based on Qingwen Bizhuo Recipe is preliminarily evaluated as superior to peracetic acid in subjective adverse reactions.
