| Objective: There are now no relevant studies of tripartite motif 21(trim21)in human non-small cell lung cancer(NSCLC)tissues analyzing its immunohistochemical expression profile.In this study,we focused on the differential expression of trim21 in NSCLC cancer tissues and adjacent normal lung tissues within 1 cm from the tumor,and analyzed and summarized the relationship between trim21 and each clinicopathological indicator.To explore the significance and potential value of trim21 in NSCLC.Methods: This experiment was approved by the ethics committee of China medicai university.A total of 132 NSCLC patient tissue samples were collected,all of which were surgically treated in the First Affiliated Hospital of China medicai university from 2019 to 2020 and the tissue samples were kept as archived cases in our department of pathology.Wax blocks containing tissues were prepared into sections following the required section criteria for immunohistochemistry,and the sections were immunostained according to the specific protocol in the experimental procedures.A scoring system was developed,and sections were scored and grouped according to their immunohistochemical expression.All clinicopathological indicators of the 132 NSCLC patients included in this experiment were statistically summarized and grouped by different criteria based on the nature of the indicators.The differences of trim21 immunohistochemical expression between NSCLC cancer tissues and adjacent normal lung tissues within 1 cm from the tumor were statistically evaluated.Each clinicopathological indicator of the 132 patients was collated and its corresponding expression scores of trim21 were counted in different groupings of each indicator,respectively.The chi square test in SPSS26.0 statistical software was used to calculate the whole data obtained in this experiment according to different indicators and grouping,to analyze the differences in the levels of trim21 in cancer tissues and adjacent normal lung tissues that are less than 1 cm from the tumor and the relationship between the expression of trim21 and each clinicopathological indicator of statistics.Results: 1.Trim21 expression was significantly attenuated in NSCLC cancer tissues.The expression of trim21 was 19.7%(26 / 132)in cancer tissues from NSCLC patients,while it was up to 90.9%(92 / 132)in adjacent normal lung tissues within 1 cm from the tumor,and the difference in expression was statistically significant(P < 0.01).2.Relationship between trim21 expression and each clinicopathological indicator.2.1 trim21 expression was correlated with NSCLC differentiation(P < 0.01),presence or absence of lymph node metastasis(P = 0.024),TNM stage(P = 0.028)and higher Ki67 expression(P =0.017).2.2 trim21 expression was not associated with age of onset,gender,drinking history,smoking history,tumor size as well as tumor typing in NSCLC patients.Conclusion: 1.Trim21 expression is significantly attenuated in NSCLC 2.The expression of trim21 is correlated with tumor differentiation,TNM stage,lymph node metastasis and Ki67 expression,suggesting that trim21 expression may inhibit the deterioration,progression and metastasis of NSCLC and serve as a new marker for diagnosis and evaluation of prognosis. |
