Objective Lung cancer is the leading cause of cancer deaths worldwide.Non-small cell lung carcinoma is the most common type of lung cancer.RSK4 acts as a protein kinase to regulate cell growth,migration,proliferation and survival.Its expression and significance in lung cancer is not clear.In this study,we detected the expression of RSK4 protein and mRNA in non-small cell lung cancer by immunohistochemistry and RT-PCR,and to explore its relationship with non-small cell lung cancer and non-cancerous non-tumor tissue expression and its relationship with clinicopathological features.To clarify the expression and significance of RSK4 in non-small cell lung cancer.Methods From Jan.2015 to Dec.2015,a total of 40 patients diagnosed with NSCLC and underwent to surgery were recruited into this study.All NSCLC diagnoses were confirmed by pathological examination,and before the experiment by the three intermediate above the pathologist physician referral no doubt.RSK4 mRNA and protein were detected by RT-PCR andimmunohistochemistry respectively.All specimens of lung cancer and non-tumor tissue were obtained during surgery.All samples of paraffin tissue were obtained from pathology.And the expression level of RSK4 gene was compared with the clinicopathological features of the patients.Results Compared with non-tumor lung tissue,the expression of RSK4 mRNA and protein in NSCLC tissues was down-regulated(P<0.01).At the same time,the content of RSK4 protein in NSCLC tissues was significantly lower than that in patients with pulmonary bullae(P<0.01).The down-regulation of RSK4 expression was associated with tumor size and TNM staging of NSCLC,but not with age,sex,CEA level,histological type of adenocarcinoma or squamous cell carcinoma.Conclusions expression of RSK4 in NSCLC tissue was down-regulated,which was correlated to the tumor size,clinical and pathological stage,suggesting that RSK4 may play a role like tumor suppressor gene in NSCLC.
