Background and Objective:Non-small cell lung cancer(NSCLC)has been the main cause of death in various cancers around the world.There have been many difficulties in early diagnosis and treatment methods.Accumulating evidence show that long non-coding RNA(lncRNA)has recently emerged as promising prognostic candidate of the cancer.LncRNA-HOTAIRM1 is frequently involved in many physiological and pathological processes of the cancer.The purpose of this study is to explore the association between LncRNA-HOTAIRM1 and clinicopathologic features.Methods:1.Quantitative Real-time PCR(qRT-PCR)method was used for evaluating LncRNA-HOTAIRM1 expression between tumor tissues and para-carcinoma tissues.2.The x2-test method was used for estimating the potential relationship between LncRNA-HOTAIRM1 and clinicopathologic parameters in NSCLC.3.The CCLE online database was performed to analyzed LncRNA-HOTAIRM1 expression in kinds of tumor cells including NSCLC.4.The Kaplan-Meier Plotter database was performed to analyze the correlation between LncRNA-HOTAIRM1 expression and overall survival(OS)in subgroups of NSCLC patients.Results:1.qRT-PCR results revealed that the relative expression of LncRNA-HOTAIRM1 in NSCLC was significantly higher(2.364±2.178 vs 1.001±0.9524,P<0.0001),compared with para-carcinoma tissues.The relative expression of LncRNA-HOTAIRM1 in 36 patients with lung adenocarcinoma was significantly higher(2.468±2.133 vs 1.073±0.8234,P<0.0001),compared with para-carcinoma tissues.The relative expression of LncRNA-HOTAIRM1 in 59 patients with lung squamous cell carcinoma was significantly higher(2.288±2.222 vs 1.001±1.037,P<0.0001),compared with para-carcinoma tissues.2.LncRNA-HOTAIRM1 was overexpressed in poorly differentiated degree of NSCLC.LncRNA-HOTAIRM1 was overexpressed in T3-T4 stages and III-IV stages of NSCLC.There was no significantly correlation between the expression of LncRNA-HOTAIRM1 and gender(P=0.5421),age(P=0.2627),histology(P=0.6595),pN stage(P=0.1367)and smoking history(P=0.2188).3.LncRNA-HOTAIRM1 was variedly expressed in kinds of malignant tumor cells by analyzing CCLE database.Expression of LncRNA-HOTAIRM1 in NSCLC was above baseline.4.The OS of lung adenocarcinoma patients with lower LncRNA-HOTAIRM1 expression was longer.In general,the OS of NSCLC patients with lower LncRNA-HOTAIRM1 expression was longer.The data showed that the OS of patients in I-II stages with lower LncRNA-HOTAIRM1 expression was longer.For smoking patients,the OS of patients with lower LncRNA-HOTAIRM1 expression was also longer.Conclusions:1.Expression of LncRNA-HOTAIRM1 was higher in tumor tissues than that in para-carcinoma tissues.2.There was significantly association between the expression of LncRNA-HOTAIRMl and histopathological differentiation,tumor size,TNM stage of NSCLC patients,but not and gender,age,histology,lymph node metastasis state and smoking history.3.LncRNA-HOTAIRM1 was variedly expressed in kinds of malignant tumor cells.LncRNA-HOTAIRM1 was expressed above baseline in NSCLC.4.Differential expression of LncRNA-HOTAIRM1 was correlated with OS of patients in NSCLC.Patients in group with low expression of LncRNA-HOTAIRM1 had longer OS than in group with high expression of LncRNA-HOTAIRM1 for lung adenocarcinoma,?-? stages of NSCLC and NSCLC with smoking history.
