| Objective By using modern biochemical technology,the hemorheology,serum PGF2_α,6-keto-pgf1_α and other indicators of primary dysmenorrhea model rats with cold coagulation and blood stasis were detected to study the therapeutic effect and mechanism of wuliji-18 wei Pill on primary dysmenorrhea of cold coagulation and blood stasis type,so as to provide more reliable theoretical basis for the clinical treatment and promotion of this prescription.Methods Sixty female Wistar rats aged 8-10 weeks were randomly divided into 6 groups:blank group,model group,Wenjing Decoction group,low-dose group,medium dose group and high-dose group of wuliji-18 pill,with 10 rats in each group.Except the blank group,the other five groups are collectively referred to as the experimental group.All the rats in the experimental group were treated with(0±1)℃ ice water bath combined with intraperitoneal injection of estradiol benzoate to prepare the primary dysmenorrhea model of cold coagulation and blood stasis type.The rats in the experimental group were divided into groups for 6 consecutive days from the fifth day of modeling.The general situation of rats in each group was observed during modeling;the writhing reaction of rats in each group was observed and recorded after medication;the uterus,ovary and kidney of rats in each group were observed and recorded The whole blood viscosity,plasma viscosity and hematocrit of rats were detected,and the levels of serum PGF2_α and 6-keto-pgf1_α were detected by enzyme-linked immunosorbent assay(ELISA).Results 1.the number of torsion reactions in the middle,high dose and Wenjing Decoction groups of wuliji-18 pill were lower than that of the model group(P<0.05);the inhibition rate of torsion in the middle and high dose groups of wuliji-18 pill were higher than that of the low dose group and Wenjing Decoction group(P<0.05).2.The organ index of uterus,ovary and kidney of rats in the middle,high dose group and Wenjing Decoction group were lower than that of model group after 6 days of treatment(P<0.05)。3.The content of serum PGF2_α in the model group was significantly higher than that in the blank group(P<0.05).The content of serum PGF2_α in low-dose group,middle dose group,high-dose group and Wenjing Decoction group was significantly lower than that in model group(P<0.05).The content of PGF2_α in middle dose group,high dose group and Wenjing Decoction group was lower than that in low dose group(P<0.05).The content of PGF2_α in middle and high dose groups of wuliji-18 wei pill was lower than that in Wenjing Decoction group(P<0.05).The content of PGF2_α in high dose group of wuliji-18 wei pill was lower than that in middle dose group(P<0.05).4.The content of serum 6-keto-pgf1_α in the model group was significantly lower than that in the blank group(P<0.05).The content of serum 6-keto-pgf1_α in model group was significantly higher than that in model group(P<0.05).The content of serum 6-keto-pgf1_αin middle dose group,high dose group and Wenjing Decoction group was higher than that in low dose group(P<0.05).The content of serum 6-keto-pgf1_α in middle and high-dose groups of wuliji-18 wei pill was higher than that in Wenjing Decoction group(P<0.05).The content of serum 6-keto-pgf1_α in high-dose group of wuliji-18 wei pill was higher than that in middle dose group(P<0.05).Conclusion 1.Wuliji-18 wei pill can inhibit the writhing reaction of primary dysmenorrhea model rats with cold coagulation and blood stasis,indicating that wuliji-18 wei pill can alleviate primary dysmenorrhea of cold coagulation and blood stasis.2.Wuliji-18 wei pill can reduce the blood viscosity of uterus in primary dysmenorrhea model rats of cold coagulation and blood stasis type,regulate the "blood stasis" state of model rats,so as to relieve the pain of model rats.3.Wuliji-18 wei pill can relieve uterine smooth muscle spasm and inhibit platelet aggregation by reducing the content of PGF2_α in serum and increasing the content of 6-keto-pgf1_α in serum,so as to treat primary dysmenorrhea of cold blood stasis type.4.The low,middle and high dose groups of wuliji-18 wei pill were used to treat primary dysmenorrhea of cold coagulation and blood stasis type.It was found that the treatment effect of middle and high dose groups was significant,and the treatment effect of high dose group was the best. |
