Study On ApoE Gene Polymorphism And Inflammatory Mediators In Zhuang Patients With Acute Coronary Syndrome

Objective:To explore the difference of ApoE gene polymorphism in ACS patients of different ethnic groups and the significance and mechanism of this difference in the pathogenesis of ACS.It provides a basis for the study of the pathogenesis of ACS patients of Zhuang nationality and the differential treatment of different ethnic groups.Methods:Included in the period from September 2018 to June 2020,ACS patients of different ethnicities who were diagnosed with ACS in the People’s Hospital of Guangxi Zhuang Autonomous Region and met the enrollment criteria(161 cases)were included in the ACS group,of which 86 were Zhuang ACS patients and 75 Han ACS patients example.The 50 patients in the control group presented with chest pain as the main complaint to our hospital and improved coronary angiography to exclude patients with coronary heart disease.The restriction fragment length polymorphism(RFLP)method is used to detect ApoE variant genes and allele frequencies.Through the application of enzyme-linked immunosorbent assay(ELISA),the serum resistin,interferon-γ(IFN-γ)and interleukin-2(interleukin-2,IL-2),interleukin-4(IL-4),interleukin-10(IL-10)level changes.Use SPSS 26.0 statistical software for analysis.The measurement data is expressed by the mean±standard deviation(x±s),and the counting data is expressed by the rate(%).Results:The PCR results of the specimens of different groups showed that in the ApoE gene in the serum of the ACS group,4 genotypes were detected in the ApoE genotype frequency,while the ε3/ε3 and ε3/ε4 genotypes were not detected.The ACS group and the control group The frequency of ApoE gene phenotype was statistically different.The ε4/ε4,ε2/ε3,and ε2/ε4 genotypes of the ACS group were higher than those of the control group,and the difference was statistically significant(P=0.032,0.000,0.000,P<0.05),but,E2/ε2 genotype ACS group and the control group were not statistically different(P=0.563,P>0.05).There are also differences in ApoE genotypes in different ethnic subgroups.The ε2/ε2 genotype of the Zhuang ACS group is lower than the Han ACS group ε2/ε2,and the difference is statistically significant(P=0.008,P<0.05),and the Zhuang ACS group ε4/Theε4 genotype was lower than the Han ACS group at the same time,the difference was statistically significant(P<0.01),the ε2/ε3 genotype Zhuang ACS group was higher than the Han ACS group,the difference was statistically significant(P<0.01),but ε2/ε4 The genotype of Zhuang ACS group and Han ACS group was not statistically different(P=0.409,P>0.05).There were also differences in inflammatory mediators between the two groups.The levels of resistin,INF-y,and IL-2 in the ACS group were significantly higher than those in the control group,and the difference was statistically significant(P<0.01),while the levels of IL-4 and IL-10 in the serum The ACS group was significantly lower than the control group,and the difference was statistically significant(P<0.01);the serum resistin and IL-2 levels in the Zhuang ACS group were lower than those in the Han ACS group,and the difference was statistically significant(P<0.01),the INF-y level in the Zhuang ACS group was higher than that in the Han ACS group,and the difference was statistically significant(P<0.01),but there was no difference in the levels of IL-4 and IL-10 between the Zhuang ACS group and the Han ACS group Statistically significant(P=0.709,0.115,P>0.05).Correlation analysis showed that the frequency of ε2/ε3 genotype in the ApoE gene was positively correlated with INF-γ;the frequency of ε2/ε2 genotype was negatively correlated with resistin levels,and the frequencies of other genotypes had no correlation with the levels of various inflammatory mediators.Conclusions:(1)The ApoE gene and its gene polymorphisms in ACS patients and non-ACS patients have different characteristics.At the same time,there are racial differences in the genotypes of Zhuang patients and Han patients in ACS patients,but the ApoE gene and its gene polymorphisms are not related to the occurrence of ACS.No obvious causality is shown.(2)The level of Th1/Th2 inflammatory mediators in ACS patients may play an important role in the pathogenesis of ACS.At the same time,the constituents of Zhuang ACS patients and Han ACS patients have ethnic characteristics and should be studied in depth.(3)Due to the limitations of research time,funding,samples,etc.,we only objectively explained the relationship between the ApoE gene and its gene polymorphisms and the characteristics of Th1/Th2 inflammatory mediators,ethnic characteristics and ACS in patients with ACS.The inner mechanism needs to be further studied.
