Objective: With Zhuang nationality as research object,this study explored the correlation between SUA and components of Metabolic Syndrome(MS)(obesity,hypertension,hyperglycemia and hyperlipidemia)as well as ZAG.Moreover,the relationship between SUA and ZAG protein was further proved by using mice with hyperuricemia as a laboratory model.This study provides a scientific basis for preventing and controlling chronic metabolic diseases and indicates a new direction for the prevention and treatment of HUA.Methods: 1.The Correlation between Biochemical Indicators of Hyperuricemia and Components of Metabolic Syndrome in Zhuang peoples of Baise(1)Those accepted physical examination of Zhuang nationality were randomized from hospitals in Youjiang district,Tiandong county,Tianyang county,Debao county,Tianlin county,Jingxi county of Baise,directly-affiliated hospital and southwest affiliated hospital of Youjiang Medical University for Nationalities.Those patients are of more than three generations of Zhuang nationality.(2)The general health indicators and biochemical indicators of subjects were tested.(3)According to the general health indicators and biochemical indicators,metabolic disorders of subjects can be classified into obesity,hypertension,hyperglycemia and hyperlipidemia.2.Analysis of Correlation between Uric Acid Level and ZAG of Zhuang peoples(1)150 patients with hyperuricemia were selected randomly from 2284 subjects of the first part.The number of subjects in healthy control group was also 150.Those subjects showed no organ diseases in heart,liver,lung,kidney and brain as well as no history of diseases in digestive tract and blood system with age from 20 to 70 years old.There are no significant differences in baseline data such as gender and age of two groups(P>0.05).(2)The expression of ZAG mRNA of subjects were detected by Real-time PCR.(3)Protein concentration in blood serum of subjects was detected by ELISA.3.Correlation between Uric Acid and ZAG in Mouse Model of Hyperuricemia(1)Mouse model of hyperuricemia was established by inhibition of uricase.(2)Serum biochemical indicators and ZAG protein concentration of mice were measured.Results:(1)Binary Logistic regression analysis: components of metabolic syndrome(overweight/ obesity,hypertension,hyperglycemia and hyperlipidemia)were independent risk factors of Zhuang peoples with hyperuricemia(P<0.05 or P<0.001).The ROC curve for hyperuricemia diagnosed by the above components were found to be statistically significant compared with 0.5(P<0.001).(2)The expression level of ZAG in blood serum of HUA group was higher than that of control group(P<0.001).Moreover,relative expression level of ZAG mRNA was rather high.Besides,there was a positive correlation between SUA and ZAG(P<0.01).The ROC curve of HUA diagnosed by ZAG was 0.770,which was significantly different from that of 0.5(P<0.001).(3)There were positive correlations between SUA and BUN,CREA,ZAG,FPG,TC,TG,LDL-C(P<0.05 or P<0.001)in mice,while HDL-C was negatively correlated to it(P<0.001).The linear regression equation of SUA and ZAG was Y(SUA)=-239.758 + 2.533 X(ZAG).Conclusion:(1)The sequence of components of metabolic syndrome of Zhuang peoples with hyperuricemia was from hyperlipidemia,hypertension,hyperglycemia to obesity which were also independent risk factors of Zhuang peoples with hyperuricemia.The diagnosis of HUA in this area was less effective.(2)There was a positive correlation between uric acid level and ZAG of Zhuang nationality.The diagnostic accuracy of hyperuricemia according to ZAG in blood serum was generally effective.(3)Mouse model of hyperuricemia can be successfully established by inhibition of uricase.The relationship between SUA and ZAG protein was further proved by using mice with hyperuricemia as a laboratory model.
