| Curcumae rhizoma is the dried rhizome of C.Phaeocaulis Val,C.Kwangsiensis S.G.Lee et C.F.Liang,and C.Wenyujin Y.H.Chen et C.Ling.The taste is pungent,bitter,warm,and returns to the liver and spleen meridians.It has the effect of eliminating accumulation and relieving pain,promoting qi and breaking blood.The commonly used forms of clinical decoction pieces include raw Curcumae rhizoma and vinegar Curcumae rhizoma.The vinegar of Curcumae rhizoma can introduce medicine into blood and enhance the effect of dispersing blood stasis and relieving pain.Literature studies have shown that there are obvious differences in appearance and chemical composition between the three kinds of Curcumae rhizoma slices and their vinegar prepared before and after preparation.However,there are differences among different slices but lacked of their own specific quality control standards.To solve the above problems,this paper choose three original raw,vinegar Curcumae rhizoma slices as the research object,using thin layer chromatography technology,fingerprint technology,component content determination technology,the electronic properties testing and evaluation of the effect of biological technology,from the"character-quality markers-biological effect"level in-depth analysis shows the quality of the similarities and differences,between different slices.By analyzing the correlation among them,a rapid identification and digital characterization method for the characters,components and biological effects of different pieces of Curcumae rhizoma was established,so as to provide a basis for improving the quality evaluation standards for the integrity and specificity of different pieces of Curcumae rhizoma.The main contents of the research are doing as follows:1.Literature researchThis paper reviewed the research progress of Curcumae rhizoma from the aspects of resources,processing,chemical composition,pharmacological action and quality standard.At the same time,the latest research progress of color measurement technology and biological effect evaluation technology is systematically summarized,which lays a foundation for the formulation of practical research program and the determination of research investigation indexes in this paper.2.Study on Quality Analysis of Different Decoction Piece of Curcumae rhizoma10 batches of samples of C.Phaeocaulis Val,C.Kwangsiensis S.G.Lee et C.F.Liang,and C.Wenyujin Y.H.Chen et C.Ling were collected,and different primitive and vinegar Curcumae rhizoma decoction pieces were prepared according to the current edition of Chinese Pharmacopoeia.Thin layer chromatography,volatile oil content determination,HPLC fingerprint and multi-component content determination were used to analyze the quality of different pieces of Curcumae rhizoma.Thin layer chromatography with reference to the current version of the"Chinese pharmacopoeia",in methadone,Curcumae rhizoma diketone,Curcumae rhizoma diketone,curcumin as contrast,petroleum ether(30~60℃)-acetone,ethyl acetate(94:5:1)for a system,spray 1%of vanillin sulfuric acid solution,in the 105℃heating to the spots were clear and the results show that different raw Curcumae rhizoma slices large difference of spectral bands,Strong specificity;Among the Germacrone ketone in three original exist,in the Curcumae rhizoma deep in the warm Curcumae rhizoma,Curdione diketone exists only in Curcumae rhizoma slices warm,shows specificity for temperature spots;The dione of Neocurdione existed only in the prepared pieces of Guangxi zedoary curcuma,which was the specific spot of Guangxi Curcumae rhizoma.Curcumin was the most obvious spot in Curcumae rhizoma.There was no significant difference in the spectrum bands between the homologous Curcumae rhizoma and vinegar Curcumae rhizoma.The preparation method(including extraction solvent,extraction method,extraction time)and chromatographic conditions(mobile phase,column,elution gradient,column temperature,detection wavelength)of the solution were investigated and optimized.The HPLC fingerprints of different Curcumae rhizoma turmeric decoction pieces were established.The fingerprint similarity was evaluated and multivariate statistical method was used to analyze the results.The results showed that the similarity of different Curcumae rhizoma was low.The PLS-DA analysis of the common peak area showed that different Curcumae rhizoma could be distinguished well.Among the same primitive and species,the similarity of Curcumae rhizoma and Curcumae rhizoma and vinegar slices was higher than 0.95,while the similarity of Curcumae rhizoma and vinegar slices from Guangxi was lower,only 0.91-0.979 and 0.901-0.973,respectively.Through PLS-DA analysis,The results showed that the quality of the decoction pieces of Curcumae rhizoma and Curcumae rhizoma was more stable than that of Curcumae rhizoma in Guangxi.The content of volatile oil in different decoction pieces of Curcumae rhizoma was determined by the method under the item of volatile oil determination in the four parts of the current edition of Chinese Pharmacopoeia.The results showed that the contents of volatile oil in Curcumae rhizoma and vinegar decoction pieces in Guangxi were1.53%and 1.48%,respectively,and the contents of volatile oil in Curcumae rhizoma and vinegar decoction pieces were 2.09%and 1.86%,respectively.The volatile oil content of Curcumae rhizoma and vinegar slices were 4.37%and 3.43%,respectively.The analysis of variance showed that the volatile oil content of Curcumae rhizoma was significantly different from that of Curcumae rhizoma.The volatile oil content of Curcumae rhizoma was reduced after vinegar preparation.Added 415 nm wavelength reference fingerprint method and determination of 9 kinds of sesquiterpenoids ingredients(Curcumenol,Neocurdione,Curdione,Isocurcumenol,Furanodienone,Curcumol,Germacrone,Furanodiene,β-Elemene)and three kinds of ingredients like curcumin(Disdemethoxycurcumin,Demethoxycurcumin,Curcumin)content,More content determination method determination of component content results show that the three original Curcumae rhizoma slices has an obvious difference,contain ingredients includi Germacrone,Furanodiene,β-Elemene in Curcumae rhizoma slices content is higher,Curcumae rhizoma Germacrone is higher in the Curcumae rhizoma slices,The content ofβ-elemene in the three primitive Curcumae rhizoma slices was low.Isocurcumenol and furanedienone only existed in Guangxi Curcumae rhizoma,Neocurdione only existed in Guangxi Curcumae rhizoma decoction pieces,curcuma dione only existed in Wen Curcumae rhizoma decoction pieces;Among the curcumin components,curcumin can be detected in both Curcumae rhizoma and,the content of which is higher in Curcumae rhizoma.Normethoxycurcumin can only be detected in Curcumae rhizoma,but dimethoxycurcumin can not be detected in all three Curcumae rhizoma,and no curcumin can be detected in Curcumae rhizoma from Guangxi.The composition changes of vinegar still need to be further studied and summarized.3.Study on the correlation between color and composition of different slices of Curcumae rhizomaThere are certain differences in the appearance color of different slices of Curcumae rhizoma,and the traditional naked eye judgment is highly subjective.Therefore,color measurement technology was adopted to digitization the appearance color of Curcumae rhizoma,and mathematical discriminant was established for rapid identification and characterization.The color characteristic data of different Curcumae rhizoma turmeric decoction pieces were collected by spectrophotometer.Through the normal distribution test,the percentile method was used to establish the bilateral 90%reference value ranges of different decoction pieces based on L*a*b*color space,and the values were respectively.The Bayes discriminant function was used to establish the mathematical discriminant model of different zedoary turmeric decoction pieces,and the commercial pieces were used to verify the results.The results showed that the positive judgment rate reached,and the established mathematical model could distinguish different zedoary turmeric decoction pieces100%.Pearson correlation coefficient was used to analyze the correlation between internal components and external color data.The results showed that curcuma dione,germalone and furandiene had significant positive correlation with L value,and isocurcumenol,β-elemene had significant negative correlation with L value.Curcumenol,furandienone,normethoxycurcumin,curcumin and B value were significantly positively correlated.4.Study on the biological potency and component index of different decoction pieces of Curcumae rhizomaAntiplatelet aggregation method was used to evaluate the blood activating biological effect of different pieces of Curcumae rhizoma.Using furandiene as the control,the"parallel line method of mass reaction"was used to calculate the blood activating potency.The results showed that the blood activating potency of Curcumae rhizoma and vinegar pieces in Guangxi were 21.2-24.8U and 17.2-20.8U,respectively.Curcumae rhizoma and vinegar slices were14.3-16.8U and 11.7-14.9U,respectively.The blood activating titers of Curcumae rhizoma were 31.2-39.7U and 23.0-28.9U,respectively.The values of vinegar Curcumae rhizoma,vinegar Curcumae rhizoma and vinegar Curcumae rhizoma were respectively.The results of variance analysis showed that there were significant differences in the biological potency of different Curcumae rhizoma decoction pieces.Warming Curcumae rhizoma had the strongest blood-promoting effect,and the biological potency of Curcumae rhizoma decreased after the preparation of Curcumae rhizoma vinegar,which was inconsistent with the enhanced effect of traditional Curcumae rhizoma vinegar on relieving blood stasis and relieving pain,and its inherent correlation still needed to be further explored.The biological potence of 10 monomer components with platelet aggregation effect was selected as the correction weight of each component.The sum of the product of content(C_i)and biological potence weight(W_i)was used to construct the blood-activating effect component index(ECI)of Curcumae rhizoma,so as to realize the comprehensive weighting of the content of effective components of Curcumae rhizoma and the biological activity of anti-platelet aggregation.By measuring only the content of components,the indexes of activating blood effect of Curcumae rhizoma can be reflected,which reduces the difficulty of evaluating the quality of traditional Chinese medicine through biological effect. |
