This paper mainly study on the best origin processing technology of Forsythiae Fructus through the chemical evaluation and biological activity evaluation,And then the chemical compositions in green Forsythiae Fructus and grown Forsythiae Fructus were analyzed and compared,so as to discuss its efficacy material base,and provide scientific basis for the rational clinical application of forsythiae.The main content is divided into the following four parts:The first part is the investigation about research situation of the origin processing of Forsythiae Fructus.This part organized the record and the research about the processing of Forsythiae Fructus,founded that the processing method of Forsythiae Fructus was disorder,and there were less research about the processing of Forsythiae Fructus.So it is necessary to adopt comprehensive assessment for the origin processing of Forsythiae Fructus.The second part is the research of the origin processing methods of Forsythiae Fructus.Based on forefathers' research,compared the streaming and boiling method.In addition,considering Chinese herbal medicine processing enterprise often stoving herbal medicine through mass production process,so different drying temperature were investigated in this study,thus process nine batch of green Forsythiae Fructus and three batch of grown Forsythiae Fructus.Taking the amount of the Forsythiae Fructus alcohol extract,the content of forsythin?rutin?quercetin?forsythiaside A?B and ?-pinene??-pinene?p-cymene?linalool as chemical evaluation index,adopted chemical evaluation of many indexes for Forsythiae Fructus.At the same time,detected the water content,total ash and the heavy metal.In addition used the activity of clear the DPPH radical and the inhibitory effect of Staphylococcus aureus and Escherichia coli as evaluation index,compare the biological activity of Forsythiae Fructus adopted different processing method.Adopted the grey pattern recognition method to evaluate the result.The result showed that there were obvious differences in different origin processing methods,the contents of ten index components in green Forsythiae Fructus are higher than grown Forsythiae Fructus.Contrast to the processed products,the raw-drying Forsythiae Fructu's content of all kinds of index are lower.Finally choose the optimum processing method of green Forsythiae Fructus was steaming 10 min,drying temperature is 60 ?,the optimum processing method of grown Forsythiae Fructus was drying in 60?.The third part is the component analysis of Forsythiae Fructus.The Non-volatile components and volatile components were researched by HPLC-MS and GC-MS method.More than 100 peaks were isolated,and 118 compounds from them were identified,the identification ratio was 96.1%;Among them the highest content was alkene compounds,account for 51.95% of the identified component,alcohol 30.55%,ketone2.61%,alkanes 2.04%,ester 1.63%,aldehyde 1.11%,acid 0.11%,heterocyclic 0.19%,epoxide0.27%.On the basis of HPLC fingerprint on by liquid chromatography-mass spectrometry,we finished components identification,24 compounds from them were identified,includes Quinic acid?Rengynic acid-1'-O-?-D-glucoside?Loganic acid?Forsythoside E?Chlorogenic acid?Caffeic acid?R-suspensaside?Secoxyloganin?Hesperidin?Rutin?Hyperin?Forsythoside A ? Plantainoside A ? Caleolarioside A ? Kaempferd-3-O-rutinoside ?(+)-Pinoresinol ?(+)-Pinoresinol--4-O-?-D-glucoside?(+)-Pinoresinol-4'-O-?-D-glucoside?Arctiin?Phillyrin?Quercetin ? Phillygenin ect.At the same time,studied the mass fragmentation rule.And provides the data foundation for the structural identification ? quantitative analysis and metabolism studies,so as to identify the compound and its derivatives quickly and accurately from the complex system.The fourth part is researching the effect of inflammatory factor released from RAW264.7 phagocytosis cells stimulated by LPS.We made the inflammation model induced by LPS in the RAW 264.7 cells to investigate the effect of Forsythiae Fructus on cells stimulated and the secretion of NO?TNF-?? IL-1? and IL-6.The results showed that the green Forsythiae Fructus and the grown Forsythiae Fructus depressed NO?TNF-??IL-6 and IL-1? secretion level in different extent,and in a dose-dependent manner,and the effect of the grown Forsythiae Fructus was superior to the grown Forsythiae Fructus.Using principal component analysis and multiple linear regression establish the ethyl acetate extract chromatogram-effect relationship on the anti-inflammatory,the results showed that the Forsythoside A was the important component.At the same time,Chlorogenic acid?Caffeic acid and Phillyrin and so on also were the anti-inflammatory component.So the anti-inflammatory effects of Forsythiae are the result of combined effect of “effective compoment gfoup.”...
