| Objective To observe the effects of different concentrations of galangin on the proliferation and migration of the A549 and PC9 cells of human lung adenocarcinoma and investigate the possible mechanisms preliminarily.Methods Lung adenocarcinoma A549 and PC9 cells were cultured in vitro,and the cells were divided into control groups(DMSO solvent control group and 0 galangin control group)and experimental groups.The experimental groups were divided according to the concentrations of galangin(12.5,25,50 and 100 μmol/L),and healthy cells at logarithmic growth stage were taken for experiments.After 24 h and(or)48 h of each group,CCK-8 experiment was used to detect the effects of different concentrations of galangin on the proliferation ability of lung adenocarcinoma A549 and PC9 cells.Cell scratch assay was used to detect the effect of different concentrations of galangin on the migration ability of lung adenocarcinoma A549 and PC9 cells for 24 h.Lactic acid assay kit,lactate dehydrogenase activity assay kit and ATP content assay kit were used to detect the effects of different concentrations of galangin on lactate production,lactate dehydrogenase activity and ATP production of lung adenocarcinoma A549 and PC9 cells.The effects of different concentrations of galangin on the expression levels of E-cadherin and N-cadherin in the supernatant of lung adenocarcinoma A549 and PC9 cells were detected by ELISA experiment.Differences between groups in each index were compared by statistical analysis.Results CCk-8 experiment showed that the cell activity of lung adenocarcinoma A549 and PC9 cells in the experimental groups was decreased to different degrees compared with the control group.In the case of the same action time of galangin,the activity of lung adenocarcinoma cells decreased with the increase of the concentrations of galangin and the difference between the groups was statistically significant compared with the control group(24h(A549: F=14.60,P<0.05;PC9: F=21.06,P<0.05);48h(A549:F=127.49,P<0.05;PC9: F=85.75,P<0.05)).In the case of the same action concentrations of galangin,the cell activity decreased with the increase of action time of galangin.Besides,the difference between the groups was statistically significant compared with the control group(P<0.05).Cell scratch test showed that the migration distance of lung adenocarcinoma cells in the experimental groups was lower than that in the control group,and the migration distance gradually decreased with the increase of galangin concentration.Besides,the difference between the groups was statistically significant compared with the control group(A549: F=141.87,P<0.05;PC9: F=223.94,P<0.05).The lactic acid test showed that the production of lactic acid in the experimental groups were lower than that in the control group.Moreover,the production of lactic acid gradually decreased with the increase of galangin concentrations.And the difference between the groups was statistically significant compared with the control group(A549: F=320.12,P<0.05;PC9: F=327.61,P<0.05).The test of lactate dehydrogenase activity showed that the experimental groups had lower lactate dehydrogenase activity than the control group.Moreover,the activity of lactate dehydrogenase gradually decreased with the increase of galangin concentrations,and the difference between the groups was statistically significant compared with the control group(A549: F=153.53,P<0.05;PC9: F=193.69,P<0.05).ATP test showed that ATP production in the experimental groups was lower than that in the control group,and ATP production gradually decreased with the increase of galangin concentrations,with statistically significant difference between the groups compared with the control group(A549: F=87.13,P<0.05;PC9: F=58.04,P<0.05).ELISA experiment showed that the expression level of E-cadherin in the supernatant of lung adenocarcinoma cells in the experimental groups was higher than that in the control group.Moreover,protein expression level gradually increased with the increase of galangin concentrations and the difference between the groups was statistically significant compared with the control group(A549: F=195.38,P<0.05;PC9: F=160.24,P<0.05).However,the expression level of N-cadherin was lower than that of the control group,besides,protein expression level gradually decreased with the increase of galangin concentrations.And the difference between the groups was statistically significant compared with the control group(A549: F=260.44,P<0.05;PC9: F=258.79,P<0.05).Conclusion Galangin could inhibit the proliferation and migration of lung adenocarcinoma A549 and PC9 cells in vitro,possibly by inhibiting aerobic glycolysis and epithelial-mesenchymal transition pathways in lung adenocarcinoma cells. |
