Objective:The paper discusses the external antitumous effect of galangin on human gliobastoma U87 and U251 cell line,the impact and action mechanism on its proliferation,invasion and migration,which provides external experimental theory basis for clinic treatment of human gliobastoma using galangin.Method:(1)cultivate human glioma cells line U87 and U251 in vitro conventionally;(2)detect the cell survival rate change after applying different concentrations of galangin(0,10,20 and 40 ?mol/L)to human glioma cells line U87 and U251 for 24h;(3)adopt wound healing experiment and Transwell experiment to detect the migration and invasion ability change of human glioma cells line U87 and U251 after applying different concentrations of galangin(0,10,20 and 40 ?mol/L)to human glioma cells line U87 and U251 for 24h;(4)adopt Western blotting method to detect TGF-? expression of human glioma cells line U87 and U251 after being processed with different concentrations of galangin(0,10,20 and 40 ?mol/L).Result:(1)CCK-8 experiment result shows that,compared with 0,10 and 20?mol/L galangin,the cell survival rate of 40 ?mol/L galangin groupdeclines(U87: F= 54.769,P= 0.000;U251: F=24.350,P= 0.000),and the difference has statistical significance(P<0.05).Galangin has a certain suppression effect on the proliferation of human glioma cells line U87 and U251,and the suppression effect appears positive correlation with the concentration.(2)Wound healing experiment result shows that after applying 0,10 and 20?mol/L galangin to U87 and U251 cell line,the ability of restraining cell migration is different.The higher the concentration is,the suppression degree is stronger(U87 cell :(47.91±2.14)%,(40.50±1.24)%and(7.36±1.45)%;and U 251 cell:(48.28±2.74)%,(40.77±2.23)% and(9.54±1.18)%)).20 ?mol/L galangin can restrain the migration ability of human gliobastoma significantly(U87 cell: F=509.321,P=0.000;U251cell: F=272.374,P=0.000),and the difference has significant statistical significance(P<0.05).(3)Transwell experiment result shows that after applying 0,10 and 20?mol/L galangin to U87 and U251 cell,the ability of restraining cell migration is different.The higher the concentration is,the suppression degree is stronger(U87 cell :66.67±21.17,216.67±20.55 and 61.67±6.43;and U251 cell : 449.33±24.17,292±8.19 and 129.33±15.01).20 ?mol/L galangin can restrain the migration ability of human gliobastoma significantly(U87 cell: F=230.027,P=0.000;U251 cell: F=262.838,P=0.000),and the difference has significant statistical significance(P<0.05).(4)Western blotting experiment result shows that after applying 0,10 and 20?mol/L galangin to U87 and U251 cell,TGF? protein expression quantity declines gradually(U87 cell: 1.00±0.00,0.42±0.04 and 0.24±0.04;U251 cell: 1.00±0.00,0.52±0.04 and 0.30±0.04).GF?protein expression of U87 and U251 cell of 20 ?mol/L galangin is lowest(U87 cell: F=564.092,P=0.000;U251 cell: F=392.261,P=0.000),and the difference has significant statistical significance(P<0.05).(5)The wound healing rate percentage of cell in TGF-?overexpression group,0?mol/L galangin group,TGF-? overexpression+20 galangin group,20 ?mol/L galangin group declines in turn(U87 cell:1.63±0.21,1.00±0.00,0.78±0.05 and 0.43±0.08;and U251 cell: 1.98±0.20,1.00±0.00,0.86±0.06 and 0.29±0.04),and the difference has significant statistical significance(P<0.05).(2)The migration quantity of U87 and U251 cell n TGF-? overexpression group,0?mol/L galangin group,TGF-? overexpression +20 galangin group,20 ?mol/L galangin group declines in turn(the migration quantity of U87 cell is 380.67±35.73,61.67±6.43,452.00±31.00 and 145.00±19.08 in turn;the migration quantity of U251 cell is 460.67±41.19,108.00±21.66,579.00±27.87 and 207.33±22.19),and the difference has significant statistical significance(P<0.05).(3)Relative TGF-? expression quantity of cell in TGF-? overexpression group,0?mol/L galangin group,TGF-? overexpression +20 galangingroup,20 ?mol/L galangin group(U87 cell: 1.00±0.00,0.43±0.08,1.63±0.21 and 0.78±0.05;U251 cell: 1.00±0.00,0.29±0.04,1.98±0.20 and0.86±0.06),and the difference has significant statistical significance(P<0.05).After overexpressing TGF-?,the suppression effect of galangin on migration and invasion of U87 and U251 cell is partly reversed,and the suppression effect of galangin on expressing TGF-? of U87 and U251 cell is partly reversed.Conclusion:Galangin can restrain the migration and invasion of human glioblastoma U87 and U251 through blocking TGF-?.
