Objective To investigate the effect of TRBP2 gene overexpression on the proliferation, migration and invasion of lung adenocarcinoma A549 cells in vitro and to study the possible mechanismMethods The human lung adenocarcinoma A549 cells were infected with the lentivirus which recombinated the human TRBP2 gene in a proper MOI and cells stably over-expressing TRBP2 were constructed by observing fluorescence intensity. TRBP2, MMP-2 and PKR mRNA expression were detected by real-time PCR; TRBP2,MMP-2 and p-PKR protein expression were detected bywestern blot. Using MTT and colony formation assay to detect proliferation. Futhermore, the transwell chambers assay and adhesion experiment were used to detectthe ability of migration, invasion, homogeneous and heterogeneous cells adhesion rate among three groups.Results The A549 cell line with the TRBP2 gene over-expressed was successfully built. Comparision with the negative control group, the invasion and migration ability of TRBP2 overexpression transfectedgroup cells were enhanced(p<0.05)and homogeneous cells intercellularadhesion rate were decreased. At the same time, adhesion rate of heterogeneous cells were increased significantly(p<0.05). The abilities ofproliferation and colony formation were enhanced (p<0.05). The expression of TRBP2 mRNA and protein were significantly raised, so wasexpression level of MMP-2 (p<0.05). The expression of PKR was no difference. however, the expression of p-PKR was significantly decreased.Conclusions By promoting the expression of MMP2 and inhibiting the phosphorylation of PKR, over-expression TRBP2 could promote the proliferation, invasion and migration of lung adenocarcinoma A549 cells.
