Study On Anti-Tumor Components Of Trillium Tschonoskii Maxim

Trillium tschonoskii Maxim.,also known as a bead on the top of the head,a traditional Chinese herbal medicine,is often used to treat high blood pressure,nerve dizziness,irregular menstruation,traumatic bleeding and other diseases.Medical research found It has anti-tumor effects.In order to further clarify the anti-tumor components in Trillium,this article uses the method of activity tracking to explore the anti-tumor components in Trillium.In this paper,the activity of Trillium was traced by normal phase silica gel column chromatography,WST-1 activity detection,liquid phase preparation and other comprehensive methods.A total of 7 samples were obtained from trillium n-butanol and the active components in the water phase.Known suspected active monomeric compounds are Chonglouoside VI(1),partialsaponin-3β-O-α-L-rhamnopyranosyl-(1→4)-[O-α-L-rhamnanopyranosyl-(1→2)]-O-β-D-glucopyranoside(2),Chonglouoside VII(3),trillenoside B(4),parispseudoside A(5),parispseudoside AR(6),26-O-β-glucopyranosyl-22α-hydroxy-(25R)-furost-5-ene-3β,26-diol-3-O-α-rhamnopy ranosyl-(1→2)-β-4-O-sulfo-glucopyranoside(7).The WST-1 method was used to detect the cell viability of the 7 suspected active monomeric compounds.The inhibitory rates of compounds 1 to 3 on He La cells at 2μg/m L were 34%,91%,and 84%,respectively.At 10 μg/m L,the inhibition rates on He La cells were 90%,97%,and 93%,respectively.The inhibitory rates of compounds5 to 6 on He La cells at 10 μg/m L were 33% and 71%,respectively.It was found for the first time that compound 5 had certain anti-tumor activity;while compounds 4 and7 had no inhibitory effect.The content of the anti-tumor active monomer compounds 1～3 in the trillium n-butanol site was determined,and the results showed that the content of compound 1was the most in the trillium n-butanol site,followed by compound 2 and compound 3,The content of compound 1～3 is 12.33%,11.52%,and 5.77% respectively.In this paper,five strong anti-tumor active monomers were obtained from the active components of Trillium by activity tracking.Among them,compound 6 is a new compound,and the anti-tumor activity of compound 5 is also reported for the first time;Trillium was established for the first time.The method for determining the content of active monomer compounds in the n-butanol site enriches the data for quality control;this article lays the foundation for the anti-tumor application research of trillium.
