Objective: To explore the mechanism of jiawei Yupingfeng(YPF)buccal tablets in the treatment of allergic rhinitis(AR)according to network pharmacology,and to study the best extraction process and quality analysis method of its active components,astragaloside,atractylenolide and polysaccharide.Methods: 1.Network pharmacology was used to predict the target and signal pathway of astragaloside and atractylenolide in AR treated with jiawei YPF buccal tablets.The network of components-target-pathway,GO Enrichment Analysis and KEGG pathway analysis of jiawei YPF buccal tablets were constructed and the main related targets and pathways were analyzed.2.the extraction effects of astragaloside,atractylenolide and polysaccharide in AR from Chinese herbs were compared by warm soaking method,ultrasonic method and water decoction method respectively.The effective extraction process for different chemical components was determined.3.The HPLC-ELSD was used to determine the content of astragaloside in the jiawei YPF buccal tablets.The content of atractylodes I,II and III were determined by HPLC.The content of total polysaccharides was determined by phenol-sulfuric acid colorimetric method under the 490 nm.The measurement method was also studied at the same time.Results: 1.27 effective components were selected from jiawei YPF buccal tablets,involving 19 target protein related to AR,including 17 targets of Astragalus,8targets of Atractylenolides,14 targets of Saposhnirovia,10 targets of Glycyrrhiza and6 targets of Peppermint.The AR related more targets of astragaloside and atractylodes were17 and 7 respectively,which was the active components for the treatment of AR with jiawei YPF buccal tablets.The main signal pathways were estrogen signal pathway,antigen processing and delivery pathway,NOD-like receptor signaling pathway and arachidonic acid metabolism pathway.Jiawei YPF buccal tablets plays an anti-inflammatory and immunoregulatory role by acting on the above targets and pathways,and at the same time induces mi RNA-21 expression by upregulating the upstream target gene,improving the Th1/Th2 ratio balance of AR and inhibiting the activation of NF-κB signaling pathway to inhibit the inflammatory response.Polysaccharide has the same effects.2.The best extraction technology ofastragaloside was warm immersion.The temperature was 75 degrees,the concentration of ethanol was 85%,and the time of warm immersion is 90 min.The extraction technology of atractylenolide was ultrasonic extraction.Polysaccharides was extracted by water decoction method.3.The content of astragaloside was determined by HPLC-ELSD.The chromatographic conditions of astragaloside were chromatographic column Diamonsil C18,the mobile phase was acetonitrile-water(34:66),volume flow rate was 1.0ml/min,The column temperature was 35 degrees,the drift tube temperature was 105 degrees,the carrier gas flow rate was 2.5L/min and the injection volume was 10 μl,there was a good linear relationship between0.416-1.664ug(r=0.9996),RSD was 1.89%(n=6).In the content determination of atractylenolide,HPLC was used to detect the atractylenolide I,II and III in the sample by wavelength switching: detection wavelength:220nm(atractylenolide I,III)and276nm(atractylenolide II).The good linear relationship between the peak area and the concentration of atractylenolide I,II and III were obtained by Y=44826X-5012.5,r=0.9999;Y=61447X-8565,r=0.9999;Y=30086X-4492.1,r=0.9999.The average recoveries of atractylenolide were 100.48%,99.85%,100.05% respectively.In the determination of polysaccharides,the relationship between absorbance and concentration with glucose as reference substance was obtained by regression equation Y=18.675X-0.0502,r=0.9998.The result show that the linear relationship between glucose concentration in 0.01-0.05ug/ml was good.The results of the above methods meet the requirements of the Pharmacopeia.Conclusion: In this paper,we used network pharmacology to predict and explore the action mechanism of jiawei YPF buccal tablets in the treatment of AR,and provided a scientific explanation for the mechanism of the treatment of AR with jiawei YPF buccal tablets from the overall perspective of biological system.The extraction process of each effective components in the prescription was reasonable,and the jiawei YPF buccal tablets were in accordance with the provisions of Pharmacopeia and the quality standard of buccal tablets was completed.The determination method and experimental data were stable and reliable.
