Objective:To observe the effect of Panax Notoginseng Injection on the changes of the intestinal microflora diversity structure in rats with chronic renal failure.Methods:Sixty SD rats were randomly divided into 6 groups: normal group,model group,low-dose treatment group,middle-dose treatment group,high-dose treatment group and benazepril group,with 10 rats in each group.Except for the rats in the normal group,the rats in each group used the 5/6nephrectomy method to establish the chronic renal falure rat model.After successful modeling,normal group and model group were given intraperitoneal injection of normal saline(10m L/kg·d);low,medium,and high-dose treatment groups were given Panax notoginseng injection 1,2,3g/kg·d,respectively Intraperitoneal injection,the benazepril group was given benazepril hydrochloride 5mg/kg·d by gavage,once a day,for a total of 8weeks of intervention.The 6 groups of rats were used to detect the renal function with an automatic biochemical analyzer;the levels of indoxyl sulfate(IS)and D-lactic acid(D-LA)were detected by ELISA;the endotoxin was quantitatively detected by the horseshoe crab test method;the bacterial populations were phylum and genus level.Results:(1)Metabolic toxin indicators: The levels of BUN,SCr,and IS in the model group increased compared with the normal group(P<0.01),but after the intervention of Sanqi injection and benazepril hydrochloride,the levels of BUN,SCr,and IS decreased The trend,the middle and high-dose treatment group and the benazepril group were lower than the model group,which was statistically significant(P<0.01).(2)The levels of endotoxin and D-LA in the model group were up-regulated compared with the normal group(P<0.01),but after the intervention of Panax Notoginseng injection and benazepril hydrochloride,each medication group had a downward trend compared with the model group Among them,the middle and high-dose treatment group and the benazepril group decreased compared with the model group,which was statistically significant(P<0.01 or P<0.05).(3)The analysis of the two abundance indexes of Ace and Chao1 showed that the abundance indexes of Ace and Chao1 in the model group decreased,which was statistically significant compared with the normal group(P<0.01).After treatment with Sanqi injection and benazepril,each The Ace and Chao1 indexes of the treatment group have a rising trend.The low,medium and high dose treatment group,benazepril group and the model group are all statistically significant(P < 0.05 or P < 0.01);Shannon diversity index analysis analysis results indicate that the similarities in each group are good.There are significant differences between the high-dose treatment group and the benazepril group and the model group and the low-dose treatment group,as well as the medium-dose treatment group,the model group and the low-dose treatment group.Significant difference,the difference is smaller than the model group and the high-dose treatment group.There are also significant differences between the normal group and the model group,but not much different from the low-dose treatment group.(5)At the phylum level,Firmicutes accounted for the highest proportion of samples in each group,with an average of more than 75%,followed by the Bacteroides and Proteobacteria.There was no significant difference between the groups of bacteria at the phylum level.(6)At the genus level,compared with the model group and the low-dose treatment group,the abundance of Enterobacteriaceae-Shigella in the benazepril group and the high-dose treatment group decreased significantly,and the lactobacillus The abundances of genus,Laurespirillum,Rumenbacterium,and Rumenococcus increased significantly.Conclusions: Panax notoginseng injection can significantly improve the renal function of chronic renal falure rats,have a significant protective effect on the intestinal mucosal barrier,and can improve the species abundance and diversity of intestinal flora.
