The Protective Effect Of Gushen Xiezhuo Heluo Recipe On The Kidney Of Diabetic Nephropathy Rats And Its Influence On Wnt/β-Catenin Pathway

Diabetic nephropathy(DN)is a kidney disease developed on the basis of diabetes,usually with a history of poor blood sugar control.As the base of diabetic patients increases,the number of people suffering from diabetic nephropathy also continues to rise.Due to diabetic nephropathy poorly control,the number of patients reaching the end point of kidney disease is also increasing year by year[1],Gushen Xiezhuo Heluo Recipe is a special prescription summed up by the tutor Director zheng yan hui in the process of treating patients with diabetic nephropathy.In the clinical process,we found that the symptoms of fatigue,edema and other symptoms of diabetic nephropathy patients after taking Gushen Xiezhuo Heluo Recipe improved to varying degrees,and related laboratory indicators proved that this prescription has a certain protective effect on diabetic nephropathy and nephropathy,but whether Gushen Xiezhuo Heluo Recipe can improve kidney pathology and based on what The mechanism of protecting the kidneys is not yet known.In this study,we studied the protective effect of Gushen Xiezhuo Heluo Recipe on the kidneys of diabetic nephropathy rats and its influence on the Wnt/β-catenin pathway.SD rats were randomly divided into normal group(N group),model group(DN group),irbesartan group(I group),Gushen Xiezhuo and Luofang low-dose group(GSL group),Gushen Xiezhuo Heluo prescription middle-dose group(GSM group),Gushen Xiezhuo and Luo prescription high-dose group(GSH group),N group and DN group were given normal saline gavage,and the remaining treatment groups were given corresponding drugs by gavage,and the administration volume was equal.It is 10ml/kg/d.The administration intervention time is 6 weeks.After the drug intervention,the rats’ blood glucose,24-hour urine protein quantification,blood urea nitrogen(BUN),and blood creatinine(Scr)were measured respectively.After the kidney was taken,the kidney tissue was sectioned for HE and PAS staining.Western blot method was used to detect expression of wnt4,β-catenin and type IV collagen.The results showed that the middle and high dose groups of Gushen Xiezhuo Heluo Recipe and the Irbesartan group can reduce blood creatinine,urea nitrogen,blood sugar,and 24-hour urine protein;HE and PAS staining can see the high dose of Gushen Xiezhuo Heluo Recipe Group,irbesartan group can significantly improve renal tubular injury and interstitial inflammatory cell infiltration,reduce matrix proliferation,improve basement membrane thickening;Gushen Xiezhuo Heluo prescription high-dose group and irbesartan group can effectively reduce the expression of wnt4 and β-catenin in the kidney tissues,and compared with the irbesartan group,the expression of wnt4 and β-catenin in the kidney tissue of the high-dose Gushen Xiezhuo Heluo prescription group further decreased;in terms of the expression of type IV collagen,only Gushen Xiezhuo Heluo prescription high-dose group effectively reducesConclusion:Gushen Xiezhuo Heluo Decoction can effectively reduce blood sugar,blood creatinine,urea nitrogen and urine protein in diabetic nephropathy rats,and reduce the expression of wnt4,β-catenin and type Ⅳ collagen,which can improve kidney pathology.The protective effect of Gushen Xiezhuo Heluo Decoction on the kidney may be related to the inhibition of wnt/β-catenin pathway reduces the overexpression of wnt4,β-catenin and type IV collagen.
