Objective:To observe the differential expression of Wnt4/β-catenin signaling pathway and Sfrp1 expression in renal tissue of diabetic rats,and to explore the role of them in thedevelopment of renal fibrosis. Methods:The 24 Sprague-Dawley(SD)rats with males were divided randomly into 3 groups: the normalcontrol group(NC) the model group(DM) and the Insulin Detemir group(INS), and was equipped with 8 in each group. One-time tail vein injection of STZ 55 mg/kg, preparation of IDDM model 72 h monitoring for three consecutive days after fasting glucose≥16.7 mmol/L. INS group were injected with 12u/kg of Insulin Detemir every 12 hours for 6 weeks. NC group and DM group was given 0.1 ml solvent,respectively. All the rats were sacrificed after 12 weeks. The pathological changes in renal tissue were detected using HE, and PAS and Masson staining.In addition,immunohistochemistry staining were employed to detect the expression of Wnt4 and β-catenin protein in renal tissue.The expression of Wnt4,Sfrp1,β-catenin, p-GSK-3β,GSK-3β,α-SMA,Ecadherin and collagen Ⅰ protein was assayed by immunohistochemical staining and western blot in rats renal tissue. Results: Compared with the NC group, the renal fibrosis were obviously in the DM group, the expression of Wnt4 m RNA and proteinwere increased significantly.(p<0.05)with decreased Sfrp1 and Ecadherin protein and increased β-catenin,p-GSK-3β, α-SMA and collagenⅠprotein(p<0.05).Compared with the DM group,the renal fibrosis was obviously relief in the INS group, the expression of Wnt4 m RNAand proteinwere decreasedaccompanied by the levels of β-catenin, p-GSK-3β,α-SMAand collagenⅠprotein expression was obvious decreased(p<0.05),theexpression of Sfrp1 and Ecadherin protein was increase(p<0.05).Conclusion:Wnt4/β-catenin signaling pathway abnormal activation in renal fibrosis of DN rats maybe relate to its inhibitory factor Sfrp1.The expression of Sfrp1 was reduced coupled with theinhibitory effect of this pathway,which was involved in the development of DN rat renal fibrosis.
