| Objective:To detect the protein expression of AXL in epithelial ovarian cancer(EOC),and analyze its correlation with clinical features and prognosis.To provide new ideas and treatment strategies for EOC targeted therapy.Method:1.Oncomine was used to watch the expression of AXL in normal ovarian tissues and ovarian cancer tissues,and Kaplan Meier was used to analyze the relationship between AXL and the occurrence,development and survival of EOC.2.From January1,2014 to December 31,2018,67 patients with EOC diagnosed by pathology in the Department of Gynecology of the second hospital of Jilin University were collected.The clinical characteristics and follow-up information of the patients were retrospectively recorded,and the paraffin sections were collected.The expression level of AXL protein in the pathological sections of 67 patients with EOC was detected by immunohistochemistry(IHC).3.Western blot was used to detect the expression of AXL protein in cancer tissues and adjacent tissues of 6 fresh EOC patients.4.SPSS 26.0 software was used.T test was used to analyze the difference of AXL protein expression in cancer and adjacent tissues of EOC patients;chi square test was used to analyze the relationship between AXL protein expression in pathological sections and clinical parameters of EOC patients;Kaplan Meier method was used to analyze the clinical data of EOC patients,the relationship between AXL protein expression in pathological sections and overall survival,and to analyze the factors affecting the prognosis of EOC patients;Cox regression analysis was used to analyze the independent risk factors affecting the overall survival of EOC patients.Result:1.The oncomine database analysis showed that AXL gene level in ovarian serous adenocarcinoma tissues was significantly higher than that in normal ovarian tissues(P < 0.05).Kaplan Meier database was used to analyze the relationship between AXL mRNA level and survival.The results showed that the overall survival and median survival of patients with low AXL mRNA level were significantly higher than those with high AXL mRNA level.2.According to the IHC results of EOC patients,the expression level of AXL protein in cancer cell membrane of high-grade EOC patients was significantly higher than that of low-grade EOC patients(P < 0.05).3.Kaplan Meier survival analysis of 67 patients with EOC showed that AXL protein was highly expressed in cell membrane(P < 0.05),EOC type II(P < 0.05),federation international of gynaecology and Obstetrics(Federation International of gynaecology and obstetrics and Gynecology)Obstetrics(FIGO)pathological stage was advanced(stage III-IV)(P < 0.05),pathological grade was high(P < 0.05),lymph node metastasis was positive(P < 0.05)were the risk factors affecting the overall survival of patients;in this study,the age of EOC patients,preoperative HE4 value,tumor size,ascites volume,preoperative CA125 were not correlated with the overall survival of patients(P > 0.05).Cox multivariate analysis showed that high expression of AXL protein in cancer cell membrane was an independent risk factor for overall survival of EOC patients(HR = 7.283,P < 0.05).Compared with eco patients with low expression of AXL protein in cancer cell membrane,the risk of death in patients with high expression of AXL protein was increased by 7.283 times.Conclusion:1.The high expression of AXL mRNA and protein is closely related to the development of ovarian cancer.High expression of AXL mRNA is a risk factor for overall survival in patients with epithelial ovarian cancer.2.High expression of AXL protein in cancer cell membrane,positive lymph node metastasis,EOC type II,FIGO stage advanced and pathological grade high are risk factors for overall survival of EOC patients.3.High expression of AXL protein in cancer cell membrane is an independent risk factor for overall survival of EOC patients. |
