| Objective: The classic recipe Danggui Buxue Decoction is selected from the51 st recipe in the "Catalogue of Ancient Classic Prescriptions(First Batch)" issued by the China Administration of Traditional Chinese Medicine.The prescription is composed of angelica and astragalus,and has the effect of nourishing qi and nourishing blood.This article studies the preparation process and quality analysis of the material standards of the classic Danggui Buxue Decoction: the original quality standards of Danggui and Astragalus are improved,and the research results are combined with modern technology research methods to determine the material standards of Danggui Buxue Decoction The preparation process is expected to provide a basis for the research and development of Danggui Buxue Decoction’s material standards and preparations.Methods: Refer to the corresponding methods under the four general provisions of the 2020 Chinese Pharmacopoeia to detect the properties,thin layer,moisture,ash,and extract of the decoction pieces;use high performance liquid chromatography to establish the characteristic map of angelica and astragalus,and determine the wine The content of ferulic acid in angelica and the content of cortisol glucoside and astragaloside IV in astragalus.In the preparation process research,the prescription dosage and preparation method are determined by consulting the records of ancient books.For the preparation process with unclear parameters,modern research methods are used to determine the effect of the decoction rate,ferulic acid,vertisol glucoside content,characteristic map and other indicators on the preparation process.The particle size,decoction,filtration,drying and other process parameters were investigated,and the preparation process of the Danggui Buxue Decoction material standard was determined on the basis of Zungu.Results: The quality standards for the decoction pieces of Chinese angelica are preliminarily formulated: the water content must not exceed 10.0%,the extract must not be less than 50.0%,the ferulic acid content must not be less than 0.07%,the total ash content must not exceed 7.0%,and the acid-insoluble ash content must not exceed 1.7%.A total of 8 common peaks were identified in the spectrum.The quality standard of astragalus decoction pieces: the moisture content should not exceed 10.0%,the extract should not be less than22.1%,the total ash content should not exceed 5.0%,the content of astragaloside IV should not be less than 0.08% based on the dry product,and the content of verbasil glucoside should not be less than 0.04 %,a total of 6common peaks have been identified in the characteristic map.The preparation process of Danggui Buxue Decoction is: 82.60 g of astragalus decoction pieces,16.52 g of distilled angelica decoction pieces,crushed two-flavor decoction pieces through a 4-mesh sieve,put them in a 2L casserole,add 800 m L of water,and use electric heating.Boil for 70 minutes(the filtrate is about400 m L),filter it with a 200-mesh filter cloth while it is hot,and freeze-dry for26h(T<-40℃,Vac<20Pa)to get the dry powder of Danggui Buxue Decoction.Conclusion: Improve the quality standards of decoction pieces and control the quality of decoction pieces from the source.The dosage of Danggui Buxue Decoction was determined according to the theory of traditional Chinese medicine and the research based on ancient records,and combined with modern research techniques,the best preparation process for the substance standard of Danggui Buxue Decoction was determined,which is the next classic recipe of Danggui Buxue Decoction.Benchmark research has laid a solid foundation. |
