Medicine.Traditional Chinese medicine compound has the characteristics of multiple components,multiple targets,and comprehensive effects.The composition of the effective substances is complex.It is the key to find an objective evaluation method that can fully reflect the quality of the compound.The effect of the compound prescription is not determined by the action of a single ingredient or the ingredient with high content,but the result of the simultaneous action of multiple index ingredients.Therefore,with multi-component analysis as the starting point,the quality,efficacy and metabolism of the compound prescription are combined to provide a theoretical reference for further research on the material basis of compound prescriptions.Research purposesIn this study,guided by TCM theories and modern analysis methods,the Chinese medicine compound Danggui Buxue Decoction was used as the research object,and the quality of the compound was evaluated from the identification of the compound’s medicinal substance,the characteristic map combined with chemometric method,and the quantitative comparison of multiple blood components.Starting from the aspect,using time-of-flight mass spectrometer and triple quadrupole liquid-mass spectrometry technology,with the help of a variety of chemical statistical analysis methods,to clarify the difference between the pathological and normal changes of the medicinal substances and multi-components of Danggui Buxue Decoction,and to nourish the blood of Angelica The identification and quality control of medicinal substances in the soup provide a theoretical basis.Research methods and results1.UPLC-Q-TOF/MS technology was used to establish the characteristic map of the Chinese medicine compound Danggui Buxue Decoction,and the characteristic map of the Danggui Buxue Decoction compound was established.The compounds of Danggui Buxue Decoction can be quickly separated within 26 minutes.The source of the main chromatographic peaks is inferred and assigned to 36 chemistries through the UNIFI platform,reference substance comparison,chromatographic retention time,molecular weight,molecular fragment peaks and related literature.The ingredients are mainly flavonoids and saponins,but also organic acids,sugars and other ingredients.Provide a theoretical basis for the follow-up study of the effective material basis of Danggui Buxue Decoction.2.Based on UPLC-Q-TOF/MS technology,11 batches of Danggui Buxue Decoction compound were measured,and similarity analysis was performed using the Chinese medicine chromatographic similarity evaluation system.The similarity of the feature maps of 11 batches was above 0.85.After the relative peak area is quantified,the common peak information is combined with the chemical recognition pattern to perform principal component analysis and cluster analysis,and 11 batches of compound prescriptions are divided into 2 categories.Through chemical recognition pattern and similarity evaluation software,an objective and comprehensive analysis and evaluation of traditional Chinese medicine compounds are carried out.3.Use cyclophosphamide combined with acetophenazine to prepare a compound blood-deficiency rat model,and give the blood-deficiency rats three doses of compound water extracts of high,medium,and low doses by gavage,and observe the rats The results showed that hemoglobin was 113 ± 13.62 g/L,white blood cell count was 4.1 ±0.18 10^9/L,red blood cell count was 6.982±0.62 10^12/L,,hematocrit was 33.48 ± 2.07%,etc.The value has been significantly improved,and the peripheral blood indicators have been significantly improved,which is statistically different compared with the model group.Taking Danggui Buxue Decoction after oral administration of normal rats and blood-deficiency rats as the research objects,a quantitative detection method was established based on liquid-mass spectrometry technology to determine four blood components ferulic acid,verbasil glucoside,and formononetin The concentration of formononetin in the body was interrupted for 11 sampling times within 0 to 720 minutes,and the drug-time curve was drawn for dynamic monitoring.Among them,the absorption of ferulic acid in model rats is faster,the peak time of formononetin in normal animals is 30 minutes,and the peak time in rats with blood deficiency syndrome is 15 minutes.Formononetin and formononetin in the two groups of animals have the same plasma concentration peak time,but the Cmax and AUC0~T are different,and the Cmax and AUC0~T of model animals are higher than those of normal animals.ConclusionGuided by the whole prescription research method,the characteristic map of the compound Danggui Buxue Decoction was established by UPLC-Q-TOF/MS technology,and the chemical components of the Danggui Buxue Decoction were separated and identified,combined with chemometric statistical methods,to preliminarily evaluate the compound quality.Under the guidance of clinical application of traditional Chinese medicine,comparing the effects of Danggui Buxue Decoction for nourishing blood and nourishing blood,the peripheral blood indicators of the blood deficiency syndrome caused by cyclophosphamide combined with acetophenazine have been significantly improved,and a quantitative detection method has been established to determine the blood in the compound.The difference in the metabolism of the ingredients in different models allows further study of the compound ingredients and mechanism of action.On this basis,it can provide a theoretical basis for the subsequent quantitative research of multiple ingredients and the elaboration of the material basis of the compound prescription. |
