| Objective:The needle knife was used to intervene the ligaments at the knee joint of rabbits with knee osteoarthritis.Observe the morphological changes of the soft tissue of the rabbit knee joint before and after the treatment,the pathological changes of the ligaments,the protein content of TGF-β1 and Smad2/3 and the m RNA expression in the ligaments,in order to clarify the recovery mechanism of the acupotomy in the KOA treatment.Needle knife treatment of KOA tissue injury provides a certain theoretical basis.Methods:Forty healthy adult rabbits were selected,male and female,weighing(2.25±0.1)kg,and divided into blank group,model group,needle knife group,and electroacupuncture group according to the random number table,with 10 rabbits in each group.Except for the blank group,they and each group were subjected to cast immobilization modeling of the left hind limbs for 6 weeks,and treatment was started the next day after modeling.In the electroacupuncture group,acupuncture was performed every other day,3 times a week,for3 weeks.Needle knife group,once a week,treatment for 3 weeks.Results:1.The general situation of rabbits: After the modeling is completed,the Leques ne MG score of the animal rises significantly,which is significantly different from the blank group,and the score is statistically significant(P<0.01).The model ani mals developed knee joint swelling,decreased joint mobility,the affected limb coul d not participate in normal walking,could not touch the bottom,kick the floor,et c.,the affected limb tenderness was obviously accompanied by a systemic reaction,and the struggle was violent.According to the morphological scoring standard anato my,it was found that the model was successful,in line with the performance of K OA.2.The protein expression of TGF-β1 and Smad2/3 in the ligaments of each gr oup(1)TGF-β1: Compared with the blank group,the expression of TGF-β1 protein in the model group was significantly decreased,and the difference was statistically significant(P<0.01);Compared with the model group,the expression of TGF-β1protein in the electroacupuncture group and the needle knife group increased to var ying degrees,and there were significant differences(P<0.01).(2)Smad2/3: Compared with the blank group,the Smad2/3 protein expression in the model group was low,and the difference was statistically significant(P<0.01);compared with the model group,the expression of Smad2/3 protein in each tre atment group was different The degree of increase,the difference is statistically sig nificant(P<0.01).3.The m RNA expression of TGF-β1 and Smad2/3 in each group ligament(1)TGF-β1m RNA: Compared with the blank group,the TGF-β1 m RNA conte nt in the ligament of the model group was significantly decreased,and the differen ce was statistically significant(P<0.01);Compared with the model group,the TG F-β1m RNA content in the ligament of the electroacupuncture group and the acupoto my group increased to varying degrees,and the differences were statistically signific ant(P<0.05).The difference between the acupuncture group and the model group was more significant.Significant(P<0.01).In addition,there was no difference bet ween the needle knife group and the blank group(P>0.05).(2)Smad2/3m RNA: Compared with the blank group,the ligament content of the model group is lower,and the difference is statistically significant(P<0.01);co mpared with the model group,the Smad2/3m RNA content in each treatment group has different degrees There are significant differences(P<0.01).4.Changes of fibers and cells in ligaments: the blank group was stained intact and did not fall off,the fibers were arranged neatly and orderly,with obvious strati fication,and the nucleus and cytoplasm were clearly visible and evenly distributed i n the fiber network.The staining of the model group showed a large area of shedd ing,the fiber arrangement was chaotic,and the fibers of each layer were chaoticall y and intersected.Only a few parts of the cell nucleus were randomly distributed i n the chaotic fiber network,and the distribution was loose.The staining of the nee dle knife group showed neat distribution of small blocks,only a small part fell off,the fibers were arranged neatly and orderly,and the layered structure was clearly visible.The cells were evenly distributed in each small block,the number was larg e,and the arrangement was also tight.Both nucleus and cytoplasm are visible.In t he electroacupuncture group,the fiber stratification can be clearly distinguished,the nucleus and cytoplasm distribution is obviously increased,but there is still some c onfusion.Conclusion:1.Both needle-knife and electro-acupuncture treatment can effectively improve knee osteoarthritis,causing benign changes in the morphology of ligaments and soft tissues,and acupotomy is more effective than the electro-acupuncture group.2.After needle-knife intervention in rabbit knee osteoarthritis,the protein expres sion of TGF-β1m RNA 、 Smad2/3 m RNA content in ligaments have changed.The t herapeutic mechanism of needle-knife may be through the signal transduction of TG F-β/Smads The regulation of the pathway is achieved. |
