Objective:To investigate the expression of mucin 1(MUC1)and β-catenin(β-catenin)in the endometrium during implantation,and to explore the role of MUC1 and Wnt/β-catenin signaling pathway in the regulation of endometrial receptivity.Methods:The endometriosis(EMs)model of SD rats was established by autologous transplantation,and the EMs rat model was intervened with gonadotropin releasing hormone antagonist(GnRH-A)and normal saline respectively,and divided into experimental group A and experimental group B,and establish a control group for surgical control.Four weeks after the successful modeling,the male and female cages were given,and the plug was picked up every morning.The negative plug was recorded as the first day of conception.On the fifth day of conception(implantation period),the SD rats in the above experiment were sacrificed and obtained.The expressions of MUC1 and β-catenin in the endometrium during implantation of the above three groups were analyzed by immunohistochemistry,Western blot(WB)and real-time fluorescent quantitative PCR.Results:1.According to the analysis of immunohistochemical technology,during the implantation period,the expression of MUC1 protein and β-catenin protein in the eutopic endometrium of experimental group A and experimental group B was higher than that of the control group,and the difference was statistically significant;the expression of MUC1 protein and β-catenin protein in the eutopic endometrium of experimental group A were higher than those of experimental group B,and the difference was statistically significant.2.According to the analysis of Western blot technology,during the implantation period,the expression of MUC1 protein in the eutopic endometrium of experimental group A and experimental group B was higher than that of the control group,and the difference was statistically significant.Although the expression level of MUC1 in experimental group A is higher than that in experimental group B,it is not statistically significant.The expression of β-catenin protein in experimental group A was higher than that in experimental group B and the control group,and the difference was statistically significant.Although the expression of β-catenin protein in experimental group B was higher than that in the control group,it was not statistically significant.3.According to the analysis of Western blot technology,during the implantation period,the expression of MUC1 and β-catenin related mRNA in the eutopic endometrium of experimental group A and experimental group B were higher than those of the control group,and the difference is statistically significant.Although the expression levels of MUC1 and β-catenin related mRNA in the eutopic endometrium of experimental group A were higher than those of experimental group B,the difference was not statistically significant.4.There is a positive correlation between the expression of MUC1 and β-catenin in endometrial tissue during implantation,which is statistically significant.Conclusion:1.Both MUC1 and β-catenin are expressed in the endometrial tissue during implantation of EMs rats and have a positive correlation.It is suggested that MUC1 may participate in the regulation of endometrial receptivity in EMs rats through the Wnt/β-catenin signaling pathway,providing a new therapeutic target for endometriotic infertility.2.GnRH-A may have an inhibitory effect on the endometrial receptivity of EMs rats.
