Objective: To explore micro-pathological mechanism of lung injury caused by dryness evil basing on rats pulmonic-tracheal morphological change and expression of SP-D and AQPs and other proteins.Methods: 60 SPF female SD rats were randomly divided into groups according to their body mass.The normal control groups(N-S and N-L)were placed in a normal temperature and humidity environment,the dry groups(D-S and D-L)and the arid groups(A-S and A-L)were placed in an artificially created dry and arid wind-sand environment for dryness evil intervention.After the intervention,the middle lobe and trachea of the right lung of the rat were collected by stages,and observed under the HE staining microscopeand used Immunohistochemical analysis of protein expression in tissues.Results: 1.Dryness evil can cause pulmonary septum thickening and congestion,alveolar fusion,small bronchial mucosal hyperplasia in the lung,focal inflammatory infiltration of tracheal mucosa with hyperemia and congestion,etc.2.The feed intake of A-S is higher than D-S(P<0.05),D-L is lower than N-L(P<0.05)and A-L(P<0.05).The water intake of N-S is lower than D-S(P<0.01)and A-S(P<0.01),D-S is lower than A-S(P<0.01),N-L is lower than D-L(P<0.01)and A-L(P<0.01).N-S is lower than D-S(P<0.01)and D-L(P<0.01),D-S is lower than D-L(P<0.01),N-S is lower than A-S(P<0.01)and A-L(P<0.01),A-S is lower than A-L(P<0.01).3.Expression of SP-D in lung N-S is lower than D-S(P<0.01)and A-S(P<0.01),D-S is also lower than A-S(P<0.01).D-S is higher than N-S(P<0.01)and D-L(P<0.01),both A-S and D-L are higher than N-L(P<0.01)and A-L(P<0.01).Expression of IL-10 in lung A-L is higher than N-L(P<0.01),D-L(P<0.01),N-S(P<0.01)and A-S(P<0.01).Expression of TNF-αin lung N-S is lower than D-S(P<0.05)and A-S(P<0.05),D-S is higher than D-L(P<0.05),A-S is higher than N-L(P<0.01)and A-L(P<0.05).Expression of AQP-5 in lung N-S is lower than D-S(P<0.01),D-L(P<0.01)and A-S(P<0.01),D-L higher than N-L(P<0.01)and A-L(P<0.01),A-S is lower than N-L(P<0.05)and A-L(P<0.05).4.Expression of SP-A in airway mucosa A-S is only lower than N-S(P<0.05)and A-L(P<0.05).Expression of SP-D in airway mucosa D-L is higher than N-L(P<0.05)and A-L(P<0.05),D-L is higher than in N-S(P<0.05)and D-S(P<0.05).Conclusions: 1.Changes in the morphology of lung tissue,trachea and bronchi mucosa,changes in the expression of SP-D,AQP-5,TNF-α,IL-10,and SP-A in lung and tracheal mucosa are microscopic manifestations of dryness evil affecting the body,the expression of SP-D and AQP-5 may be closely related to the defense function outside the lung guard.2.The micro-pathological mechanism of lung injury caused by autumn dryness evil and the combination of dry,fire,wind and cold are same but different.
