| Objective:Network pharmacology and bioinformatics were used to predict the molecular mechanism of Qiangzhi prescription in the treatment of Tourette syndrome(TS),and to evaluate the clinical efficacy and safety of Qiangzhi prescription.Methods:1.In the treatment group,30 cases were treated with oral administration of Qiangzhi prescription Avoid decoction granule.In the control group,30 patients were treated with oral Tiapride for 12 weeks.YGTSS Yale scale and TCM syndrome score scale were used to score the children before and after treatment to evaluate the clinical efficacy and safety of the two groups.2.Network pharmacology research methods: network pharmacology and bioinformatics were used to predict the potential targets of Qiangzhi prescription,and TS target genes were searched through public database to identify specific targets of Qiangzhi prescription to treat TS,and differential expression genes of TS were mined through GEO chip.Results: 1.Clinical observation results: compared with the control group,there were statistically significant differences between the treatment group and the control group in the number of twitches,frequency of twitches,and therapeutic effect of TCM syndrome score(P<0.05).In terms of YGTSS total score,motility twitch,vocal twitch,complexity twitch intensity,and impact on life or behavior,there was no significant difference between the treatment group and the control group(P>0.05),indicating that the therapeutic effect was similar between the treatment group and the control group.The adverse reaction rate in the treatment group was lower than that in the control group(P<0.05),and the difference was statistically significant.2.Results of network pharmacology research: 61 active ingredients and785 targets in Qiangzhi prescription were screened out through database query.In the GO enrichment analysis,the top 20 were selected according to the False discovery rate(FDR)for key analysis: there were 7 molecular function items,7 cellular component items and 11 biological process items.In the KEGG pathway analysis function,9 pathways were screened according to FDR< 0.05,among which the smaller FDR pathway was the interaction stimulating nerve tissue,and a total of 53 genes were enriched in this pathway.Conclusion:The treatment of TS with Qiangzhi prescription is effective and the adverse reaction rate is low.Qiangzhi prescription may through the dopamine system,adrenergic system,serotonin system,MAPK/ERK cascade system,monoamine oxidase A multiple target multiple pathways,such as role in cell proliferation are regulated,nerve mother is regulation of cell proliferation,the activity of adenylate cyclase negative regulation,renin receptor signal transduction pathways,including making RAC1,RAB7 A,KDM4B,SLC5A3 cut,the expression of intervention control Tourette syndrome. |
