Observation Of The Clinical Efficacy Of WenjingTang In The Treatment Of Yang Deficiency And Cold Coagulation And Blood Stasis PCOS And The Study Of The Mechanism Based On Network Pharmacology

Objective:The clinical efficacy of WenjingTang in the treatment of polycystic ovarian syndrome(PCOS)in the treatment of yang deficiency and yang deficiency and cold coagulation stasis was observed,and the active ingredients,targets,and pathways of WenjingTang in the treatment of PCOS were analyzed through network pharmacology,and its mechanism of action was discussed,which provided a theoretical basis for further research and clinical application.Methods:1.From August 2020 to October 2021,he was diagnosed as a patient with cold coagulation and blood stasis type PCOS in the Gynecology Clinic of the Affiliated Hospital of Chuanbei Medical College for the first time,and collected the data of the menstrual cycle,TCM syndrome score,BMI,endocrine indicators and other data before and after treatment,and conducted research and analysis.2.Download the GSE5090 dataset from geodatabase to calculate and analyze PCOS differential genes,excavate the chemical composition and action targets related to WenjingTang,build a compound regulation network for WenjingTang treatment of PCOS,screen the core targets of WenjingTang treatment PCOS by constructing the protein interaction network between WenjingTang and PCOS targets,and finally conduct GO and KEGG enrichment analysis on disease and drug intersection targets.Results:1.After the end of 1 course of treatment,3 cases were cured,6 cases were effective,and 21 cases were effective,with a total effective rate of 88.2%.After treatment,the patient’s menstrual cycle,TCM symptoms,and sex hormone levels decreased compared with before treatment(P<0.05),and the clinical efficacy before and after was statistically significant.2.A total of 218 active ingredients of WenjingTang were obtained,and 22 corresponding rake points and PCOS intersection rakes of the active ingredients were obtained,and the core active ingredients such as glycine,naringin,quercetin,and kaempferol and core targets such as VCAM-1,CTNNB1,and AKT1 were screened.A total of 267 BP items,4 CC items,and 52 MF items were obtained by GO enrichment analysis,mainly related to nutritional reactions,ketone metabolism,antioxidant reactions,etc,and KEGG signaling pathway enrichment analysis showed 10 pathways,mainly involving AGE-RAGE signaling pathway,AMPK signaling pathway,shear force,and atherosclerotic signaling pathway.Conclusions:1.After 1 course of oral treatment of WenjingTang in patients with yang deficiency and cold coagulation stasis PCOS,it can effectively improve menstrual cycle disorders,renal deficiency,and blood stasis,and reduce serum androgen levels.2.The active ingredients of WenjingTang have anti-inflammatory,antioxidant,reduce insulin resistance and other effects,by activating the AMPK signaling pathway,inhibiting the release of inflammatory factors,adjusting the body’s energy metabolism,regulating the production of gonadal steroid hormones,alleviating insulin resistance,thereby playing a role in the treatment of PCOS.
