| Objectives:This experiment through the establishment of T2DM model mice,Xiehuo Jiedu Yangyin Recipe(XHJDYY)is combined use of classical formula,to investigate the effects of different dosages of XHJDYY on LPS-TLR4/NF-κB pathway in type 2 diabetes model mice and the optimal dosages.Methods:60 SPF male C57BL/6 mice were randolly divided into 6 groups:normal group,model group,metformin group and traditional Chinese medicine group(high、middle and low dose group).The model group were produced by intraperitoneal injection of one-time with STZ.After 4 weeks of treatment,fasting for 16h after the last administration,body weight and blood glucose were measured,HE staining to observe liver and pancreatic lesions,serum levels of Il-6 and Il-1β were detected by ELISA,and protein expression levels of TLR4,MyD88 and NF-kB were detected by Western blot.Results:After 4w administration,pancreas and liver disease improved compared with model groupthe,blood glucose in the traditional Chinese medicine group and the metformin group decreased(p<0.01),the body weight increased(p<0.01),the expression levels of TLR4,Myd88,and NF-kB decreased(p<0.05),and the levels of IL-6,IL-1β in the metformin group and the medium and low dose groups of XHJDYY prescription decreased significantly(p<0.05).The comparison between the traditional Chinese medicine group and the metformin group was basically the same(p>0.05).Conclusion:The mechanism may be related to the inhibition of the inflammatory signaling pathway of LPS-TLR4/NF-kB and the down-regulation of the expression levels of inflammatory factor IL-6,IL-1β and NF-B,MyD88 and TLR4 in renal tissues. |
