Objective:On the basis of previous experiments,the effects of Xiezhuo Jiedu Recipe on the contents of Lactobacillus,Bifidobacterium,Enterococcus and Enterobacter in feces and the expression of IL-10 and TLR4 in serum of UC model rats after treatment and after secondary modeling were observed.To explore the possible mechanism of Xiezhuo Jiedu Recipe in preventing UC recurrence,and to provide strong scientific basis and guarantee for clinical application.Methods:100 healthy and clean Wistar rats were selected,randomly selected 10 rats as blank group(N),the rest rats establish UC model by 80mg·kg~-11 TNBS 50%ethanol.After confirming the success of the moulding,the remaining rats were randomly divided into Model group(M),sulfasalazine group(S),low(ZL),medium(ZM)and high(ZH)dose groups of Xiezhuo Jiedu Recipe,17 rats in each group.All groups except group N were given corresponding drugs by gastric lavagel.After fourteen days,five rats in the blank group and eight in each treatment group were randomly executed.HE staining for histopathological changes of colon.The expression of TLR4 in colon tissues was observed by immunohistochemistry.ELISA was used to detect IL-10 in serum,and q-PCR was used to detect the relative content of four bacteria in feces.The remaining rats were fed with 30 mg·kg~-11 TNBS50%ethanol for 2 days,and then all the rats were killed and the detection index is the same as the above.Result:1.The general condition:Compared with group N,rats in group M were in poor condition with mucus and pus in their stools.After 14 days,the general condition of rats in each group was improved.After the second modeling,the disease recurred in the M,S and ZL groups.The general condition was similar to that after the first modeling.The general condition of the ZH and ZM groups was good.2.Disease Activity Index(DAI)Score in RatsCompared with group N,DAI scores of rats in each group increased significantly(P<0.05).Compared with group M,the scores of other treatment groups were lower(P<0.05).Compared with group S,ZH group decreased significantly(P<0.05).After the second modeling,the score of S and ZL group was higher than that of corresponding treatment(P<0.05).There was no significant difference among M,ZM and ZH groups(P>0.05).3.Colonic mucosal index(CMDI)scoreCompared with group N,the scores of CMDI in each group were significantly higher(P<0.05).Compared with group M,the scores of other treatment groups were lower(P<0.05).Compared with group S,ZH group decreased significantly(P<0.05).After the second modeling,the score of S and ZL group was higher than that of corresponding treatment(P<0.05).There was no significant difference among M,ZM and ZH groups(P>0.05).4.The level of IL-10 of serum in ratsCompared with group N,the level of IL-10 in serum of rats in each group decreased significantly(P<0.05).Compared with group M,the rest of the treatment group increased(P<0.05).Compared with group S,ZH group increased significantly(P<0.05).After the second modeling,the content of S and ZL decreased compared with the corresponding treatment(P<0.05).There was no significant difference among M,ZM and ZH groups(P>0.05).5.The expression of TLR4 in Colon Tissue of RatsCompared with group N,the expression of TLR4 in colon of rats in each group increased significantly(P<0.05).Compared with group M,the expression in other treatment groups decreased(P<0.05).Compared with group S,ZH group decreased significantly(P<0.05).After the second modeling,the expression of S and ZL increased compared with the corresponding treatment(P<0.05).There was no significant difference among M,ZM and ZH groups(P>0.05).6.The contents of four bacteria in rat fecesCompared with group N,Lactobacillus and Bifidobacterium in feces of rats in each group decreased significantly,while Enterobacter and Enterococcus increased(P<0.05).Compared with group M,the former two decreased significantly in the remaining treatment group,while the latter two increased(P<0.05).Compared with group S,the former two increased significantly in ZH group,while the latter two decreased(P<0.05).After the second modeling,compared with the corresponding treatment,the former two groups decreased significantly,while the latter two increased(P<0.05).There was no significant difference among M,ZM and ZH groups(P>0.05).7.Analysis of correlation between IL-10,TLR4 and four kinds of bacteriaIL-10 was positively correlated with Lactobacillus and Bifidobacterium,but negatively correlated with Enterobacter and Enterococcus(P<0.05).TLR4 is the opposite.Conclusion1.In UC model rats,the level of IL-10 in serum decreases,TLR4 in colon tissue increases,and intestinal flora imbalance exists.2.Xiezhuo Jiedu Recipe can improve the general condition of UC rats,decrease the damage score of colonic mucosa,increase the content of IL-10,decrease the expression of TLR4,regulate the proportion of beneficial and harmful bacteria in intestine.It is an effective method to treat UC.3.The mechanism of Xiezhuo Jiedu Recipe in preventing the recurrence of ulcerative colitis may be through regulating intestinal flora,increasing the content of IL-10 and reducing the expression of TLR4,thereby enhancing the immune regulation.
