Study On The Distribution Characteristics Of Tcm Constitution In Metrorrhagia And Metrostaxis And The Evaluation Of The Animal Model Of Blood Stasis And Heat

Objective:To explore the correlation between metrorrhagia and metrostaxis(Ovulatory abnormal uterine bleeding（AUB-O）and constitution of traditional Chinese medicine;At the same time,to evaluate the Rat Model of Blood Stasis and Heat.Methods:First,We established a TCM Constitution uestionnaire of the metrorrhagia and metrostaxis,then,launched the clinical investigation,last,statistically analyzed the characteristics,influencing factors of this disease by using the methods of frequency analysis、chi-square test.The experimental research based on the SD female infertile rats as subjects.We evaluated the advantages of rat AUB-O model of Blood Stasis and Heat.Result:1.In the clinical investigation of the metrorrhagia and metrostaxis,42-48 years old was the age vulnerable;The occupations of mental worke accounted for the majority;The normal body mass index accounted for the majority;Pregnancies less than one was in the majority;The majority of births was one;The mode of production was mainly spontaneous delivery;Women with regular menstruation was in the majority;Two kinds of constitutions were common（44.55%）;According to frequency statistics,Qi deficienc（20.18%）,Qi depression（18.50%）,Yin deficiency（13.46%）and damp-heat（12.23%）were common.Qi deficiency were often accompanied with blood stasis,Qi depression were often accompanied with damp-heat.Through the Single factor analysis,we can get the conclusion that the number of pregnancies was the influencing factors of the time of vaginal bleeding,rather than age,BMI,occupation and contraceptive methods;Hemorrhagic pattern of this disease was related to TCM constitution.Metrorrhagia was closely related to damp-heat and blood stasis,metrostaxis was closely related to normal-constitution、Yan deficiency、Qi depression;Metrorrhagia and metrostaxis was closely related to normal-constitution、Yan deficiency、Qi depression、special constitution;2.AUB-O model of Blood Stasis and Heat and Estrogen had no significant difference in the expression of estrogen（E₂）and progesterone（P）;There was no change of estrous cycle and pathological changes were in proliferative stage;However,the rats in AUB-O heat-stasis group showed signs of pyogenic secretion,withered hair,livid tail and other symptoms of heat-stasis in general morphology.The expression of TXA₂ and IL-1 in AUB-O heat stasis group were significantly higher than those in estrogen group,while the expression of PGI₂ and TGF-βwere significantly lower than those in estrogen group.The whole blood viscosity and plasma viscosity increased significantly in AUB-O heat stasis group,and the uterine congestion and edema was obvious in this group;The results suggested that the intima of rats after gavaging estrogen combined with non-invasive placement of copper clad rod shows proliferative changes,also have characterization of Stasis-heat Syndrome and Changes of Physical and Chemical Indicators,which conforms to the clinical characteristics of this disease.It is a good animal model for studying the pathogenesis and prevention mechanism of Stagnant-heat metrorrhagia and metrostaxis.Conclusion:The clinical investigation showed that qi deficiency and qi depression were the main constitutions of the disease,and the hemorrhage pattern was related to constitution,and the“blood stasis and heat”played an important role in the pathogenesis of the disease.Experiment research proved that AUB-O model of Blood Stasis was more suitable to describe the clinical process of the characteristics of the metrorrhagia and metrostaxis.
