| Objective:The quantitative analysis of Ferrous ion(Fe2+),Ferric ion(Fe3+),phenolic acid from processed products of scrap iron;and pharmocodynamic on Iron Deficiency Anemiac by determining the concentration of hemoglobin,serum iron and hepatic iron.All of them are aim to clearly elucidated the mechanism of processed methods.Methods:Firstly,prepared processed iron products of decocting method and immersion method;secondly,determined the contents of totally iron,Fe3+and Fe2+by using Iodometric,EDTA titration methods and Ultraviolet-visible spectrophotometry;determined the contents of phenolic acid by using HPLC;Lastly,established rat model of iron deficiency anemia by feeding low iron diet combined with tail vein bleeding,adopted Seven-flavor iron pills,processed products of iron,and Ferrous Succinate Tablets to cure IDA;Results:(1)The content of total iron in the raw scrap iron was 83.24%,the contents of Fe3+and Fe2+were 29.88%and 0.0005%,respectively.(2)Terminalia chebula as an adjunct,the content of Fe3+decreased to 24.05%and26.35%,and the Fe2+increased to 2.46%and 0.92%,respectively.(3)The raw Terminalia chebula contained 1.67%for gallic acid,2.23%for corilagin,9.33%for chebulagic acid,1.18%for ellagic acid and 24.70%for chebulinic acid,and the content of phenolic Acid decreased obviously after processing.The contents of gallic acid were 1.24%and 1.51%,respectively,and the contents of the other acid were reduced to 0%.(4)Using gallic acid to instead of Terminalia Chebula as adjunct,the content of Fe3+in decocting and immersion methods were reduced to 26.42%a nd 28.44%,and the Fe2+was increased to 0.33%and 0.14%,respectively.The reaction product of gallic acid with iron was identified as pyrogallic acid by separation and identification.(5)According to the requirements of the Chinese Pharmacopoeia in 2015 to prepare Seven-flavor iron pills by means of processed products of iron.According to the requirements of Chinese Pharmacopoeia,microscopical identification and thin layer chromatography were used to identify the Aucklandiae Radix,Dracocephalum tanguticum and Carthamus tinctorius in the prescription.The content of costundide in the prescription was determined by HPLC,and the results were in accordance with the Pharmacopoeia standard.(6)A rat model of iron deficiency anemia was established by using low iro n diet combined with taking blood from tail vein.The hemoglobin content was reduced to86.35 g/L,and the hemoglobin content of normal group was 105.59 g/L,respectively,there was a significant difference in hemoglobin and serum iron of two groups(P<0.01).After 2 weeks of treatment,the hemoglobin and serum iron contents in the model group were recovered,and the recovery hemoglobin and the serum iron contents in the Ferrous Succinate tablets group were 93.84 g/L and 2.01 mg/L,respectively.Others:the contents of compound Seven-flavor iron pills and processed products of iron were 109.76 g/L and 109.94 g/L in low dose,112.39 g/L and 113.08g/L in medium dose,respectively.The therapeutic effect of compound and iron scrap alone was the same(P>0.05),but better than that of Ferrous Succinate(P<0.01).The hemoglobin content of compound and iron were 96.98 g/L and 95.12 g/L,respectively,and the therapeutic effect was equivalent to Ferrous Succinate(P>0.05).Recovery of serum iron:The therapeutic effects of in compound Seven-flavor iron pills low dose was 4.22 mg/L and 3.22 mg/L,and in medium dose,at 3.28 mg/L and 3.19 mg/L 3.22mg/L,was better than those in Ferrous Succinate(P<0.05).At high doses,the concentrations were 2.69 mg/L and 2.26 mg/L,respectively.Conclusion:(1)Terminalia chebula could reduce Fe3+to Fe2+.The gallic acids in scrap iron can play the role of antioxidants,prevent Fe2+from being oxidized to Fe3+.(2)The scrap iron processed by Terminalia chebula had a good effect on the reco very of low hemoglobin and serum iron level in iron deficiency anemia,and the effect was better than that of Ferrous Succinate(P<0.05).The effects of compound Seven-flavor iron pills and single iron processed products were similar at the same dose,sho wing a dose-dependent manner,but could not be overused,resulting in a reduced effect. |
