| Objective: The prevalence of obesity and its associated metabolic abnormalities is increasing significantly worldwide,increasing the risk of many diseases such as cardiovascular disease,type 2 diabetes,hyperuricemia,nonalcoholic fatty liver disease,respiratory diseases such as Corona Virus Disease2019 and even cancer.Fat mass and obesity-associated gene(FTO)are the genes most strongly associated with obesity.The gene of ATP-binding cassette subfamily G member 2(ABCG2)encodes urate transporter,in which Q141 K is the most significantly correlated locus with HUA in Asian population.Therefore,we will analyze :(1)The effects of obesity on glucolipid and uric acid metabolism parameters;(2)The correlation between rs9939609 and rs17817449 polymorphism of FTO gene and obesity was verified repeatedly,and the relationship between Q141 K polymorphism of ABCG2 gene and obesity was investigated;(3)Whether rs9939609 and rs17817449 polymorphisms of FTO gene and Q141 K polymorphisms of ABCG2 gene have effects on glucolipid and uric acid metabolism parameters,respectively.Methods: A total of 376 patients who underwent physical examination in Huangjiahu Hospital from April 2021 to June 2021 were selected.According to age and body mass index(BMI),the participants were divided into: Total cohort(Overweight/obesity group,n=193;Normal weight group,n=183),age range28-68 years;Youthful cohort(Overweight/obesity group,n=74;Normal weight group,n=107),age range 28-40 years;The middle-aged and the elderly cohort(Overweight/obesity group,n=119,Normal weight group,n=76)had an age range of 41-68.The effects of overweight/obesity on metabolic parameters were analyzed by comparing glucose(Glu),total cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL),low density lipoprotein(LDL)and uric acid(UA)levels among all groups.Then,the genotypes of rs9939609 and rs17817449 of FTO gene and Q141 K of ABCG2 gene were analyzed to investigate whether the polymorphisms of these loci were associated with overweight/obesity and metabolic parameters.Results: The metabolic parameters of overweight/obesity group were significantly different from those of control group.The rs9939609 and rs17817449 polymorphisms of FTO gene were strongly correlated with overweight/obesity.Haplotype analysis showed that haplotype GA and GT had2.99 and 1.81 folds risk of overweight/obesity,respectively.The Q141 K polymorphism of ABCG2 gene was more correlated with obesity than FTO gene.The risk alleles of rs9939609,rs17817449 and Q141 K increased the risk of abnormal metabolic parameters,and haplotype GT had a strong influence on the levels of TC,TG and HDL,while haplotype GA had a stronger correlation with TG and HDL.Conclusions: In this study,we verified the role of rs9939609 and rs17817449 risk alleles and haplotypes of FTO gene in overweight and obesity,and proved for the first time that Q141 K polymorphism of ABCG2 gene was associated with obesity.We also found that the risk allele(A)of rs9939609 was associated with the risk of high TC,high TG,low HDL and high UA level,and the risk allele(G)of rs17817449 was associated with the risk gene of high TG and low HDL.GT and TT genotypes at Q141 K of ABCG2 gene have extremely significant effects on the increase of UA level,and TT genotypes also have important effects on high TG and low HDL.The mechanism of dysregulation of glucolipid and uric acid metabolism parameters is complex.The introduction of FTO,ABCG2 gene polymorphism into the known single nucleotide polymorphism(SNP)of metabolic disorders is beneficial to evaluate the risk of metabolic disorders in people carrying more risk alleles,and provides a new basis for clinical prevention and individualized treatment of hyperlipidemia and hyperuricemia. |
