Objectives To study the correlation between MTHFR gene polymorphism and ABCG2 gene polymorphism and stroke.A deeper understanding of the pathogenesis of stroke provides a theoretical basis for precision medicine and primary prevention of stroke.Methods 1 Research object A total of 184 stroke patients admitted to the affiliated hospital of north China university of science and technology from November 2018 to October 2019 were randomly selected to be included in the stroke group.During the same period,184 healthy patients in the affiliated hospital of north China university of science and technology were randomly selected to be included in the control group.2 Research methods the baseline data collection accurately collects the general baseline data of the two groups of subjects.Age,sex,height,weight,history of hypertension,smoking,alcohol use,diabetes,family history and stroke were included,and biochemical results:blood lipid series,uric acid levels,homocysteine values.Fasting peripheral blood was collected and cryopreserved.Extraction of DNA from experimental subjects,qualitative and quantitative analysis of MTHFR polymorphism and ABCG2 polymorphism by real-time PCR.3 Statistical analysis was carried out by using SPSS 22 edition statistical software.The statistical data were expressed by,the significance of the difference between the two groups was analyzed by t test,the counting data were expressed by frequency,and the significance of the difference between the two groups was evaluated by 2 test.Hardy-Wilhelm Weinberg's law is tested by the standard goodness-of-fit Test 2,and hardy-Wilhelm Weinberg's law is analyzed by the standard goodness-of-fit test 2,the allele frequency and genotype frequency of stroke group and Control Group were analyzed by 2 test.Binary logistic regression analysis was used to analyze the correlation of risk factors of stroke.P<0.05 the difference was statistically significant.Results 1 MTHFR gene polymorphism:There was no significant difference in the actual and theoretical frequencies of each genotype between the stroke group and the control group(P>0.05),which was consistent with the“Hard-weinberg law”.There was a statistically significant difference in genotypes between the stroke group and the control group(P<0.05),and there was a statistically significant difference between the C and T alleles between the two groups(P<0.05),suggesting that the polymorphism of MTHFR gene rs1801133 may be correlated with stroke.The heterozygous CT type in the two groups was compared with the wild CC type,suggesting that the risk of stroke in the heterozygous CT type was 1.343 times that of the wild CC type(P<0.05);The comparison between TT type and wild CC type in the two groups suggested that TT type was 1.914 times of the risk of stroke compared with wild CC type(P<0.05).2 ABCG2 gene polymorphism:There was no significant difference in the actual and theoretical frequencies of each genotype between the stroke group and the control group(P>0.05),which was consistent with the“Hard-Weinberg law”.There was a statistically significant difference in genotypes between the stroke group and the control group(P<0.05),and there was A statistically significant difference in alleles A and C between the two groups(P<0.05),suggesting that polymorphism at rs2231142 of ABCG2 gene may be correlated with stroke.The comparison of heterozygous AC type between the two groups and wild CC indicated that the risk of stroke of heterozygous AC type was 1.568 times that of wild CC type(P<0.05).The comparison between the two groups with the mutated AA type and the wild CC type suggested that the risk of stroke with the mutated AA type was 4.471 times that of the wild CC type(P<0.05).3 Risk factors of stroke:binary logistic regression:Heterozygous CT type and mutated TT type at rs1801133 of MTHFR gene increased the risk of stroke compared with wild CC type(P<0.05);The heterozygous AC type and mutation AA type at rs2231142 of ABCG2 gene increased the risk of stroke compared with wild CC type(P<0.05);Smoking,alcohol consumption,hypertension,diabetes mellitus and homocysteine were risk factors for stroke(P<0.05).Conclusions 1 Polymorphism at rs1801133 of MTHFR gene may be correlated with stroke,and heterozygous CT type and mutated TT type may increase the risk of stroke.2 Polymorphism at rs2231142 of ABCG2 gene may be correlated with stroke,and heterozygous AC type and mutation AA type may increase the risk of stroke.Figure8;Table10;Reference 110...
