Regulation Of Shenling Baizhu Powder On 5-HT Of Brain-gut Axis In Rats With Spleen Deficiency And Diarrhea

Spleen deficiency and diarrhea is a common clinical disease in traditional Chinese veterinary medicine,which can cause high mortality in young animals and very harmful to the breeding industry.Shenling Baizhu Powder is a classic prescription for the treatment of spleen deficiency and diarrhea by traditional Chinese veterinarians.Pharmacological studies have shown that the curative effect of Shenling Baizhu Powder on spleen deficiency and diarrhea syndrome is related to its effects on enhancing body immunity,protecting digestive tract mucosa,regulating intestinal microflora,etc.,but studies on its regulating effect on brain-gut axis 5-HT less.Therefore,this experiment established a rat model of spleen-deficiency diarrhea,starting with the effect of Shenling Baizhu Powder on the amount of 5-HT secretion in the intestine and brain of rats with spleen-deficiency diarrhea,and exploring its regulatory effect on the brain-gut axis.In this experiment,40 male SD rats were randomly divided into control group,model group and Shenling Baizhu Powder high（20 g/kg）,medium（10 g/kg）,and low（5 g/kg）dose groups,using"The diarrhea model of spleen deficiency and diarrhea in rats was established by the method of“bitter cold diarrhea,eating disorder and excessive fatigue”,and treated with different doses of Shenling Baizhu Powder.Observe the general condition of the rat,histological changes in the intestines,combined with changes in body weight,diarrhea index,AMS and D-xylose content,to evaluate the effect of model building and the therapeutic effect of Shenling Baizhu powder;serum was measured with an ELISA kit 5-HT content,q RT-PCR method to determine the relative expression of jejunum TPH-1,SERT,5-HT₃R,5-HT₄R m RNA,immunohistochemical method to examine the expression of 5-HT in the hypothalamus and midbrain,and discuss the treatment of Shenling Baizhu Powder The effect of spleen deficiency and diarrhea on the 5-HT metabolic pathway of the brain-gut axis.The results showed that the rats in the model group were fatigued,lethargic,thin stools,severely shed intestinal villi,disorderly arranged,and increased lymphocyte numbers;compared with the control group,weight loss（P<0.01）,AMS and D-wood The sugar content decreased and the diarrhea index increased significantly（P<0.01）;compared with the model group,the rats in the middle-dose Shenling Baizhu Powder group had better mental status,significant weight gain（P<0.01）,and intestinal villi were basically intact and arranged neat,the diarrhea index was significantly reduced（P<0.01）,and the content of AMS and D-xylose increased.The results of ELISA,q RT-PCR and immunohistochemistry showed that compared with the control group,the serum 5-HT content of the model group increased significantly（P<0.01）,and the relative expression of TPH-1 and 5-HT₃R m RNA in the jejunum increased High,the relative expression of SERT and 5-HT₄R m RNA was significantly reduced（P<0.01）,and the positive expression of5-HT in the hypothalamus and midbrain was significantly reduced（P<0.05）;compared with the model group,the serum 5-HT content of rats in the middle dose of Shenling Baizhu Powder group was significantly decreased（P<0.05）,the relative expression of TPH-1 and 5-HT₃R m RNA in the jejunum decreased,the relative expression of SERT and5-HT₄R m RNA increased,and 5-HT in the hypothalamus and midbrain The positive expression was significantly increased（P<0.05）.In conclusion,the middle dose（10 g/kg）of Shenling Baizhu Powder has a good therapeutic effect on experimental rats with spleen deficiency and diarrhea syndrome,its mechanism of action may be related to regulating gastrointestinal function and regulating the expression of 5-HT in the brain-gut axis quantity related,the hippocampal expression level is not significantly different（P>0.05）.
