Spleen deficiency diarrhea is one of the common diarrhea syndromes in animal production,which is caused by spleen deficiency leading to abnormal circulation and stagnation of water and dampness,seriously affecting the development of the aquaculture industry.In the context of antibiotic prohibition,traditional Chinese medicine has become a promising alternative drug due to its advantages such as low cost,no drug resistance,and no residual chemical drugs.Qiwei Baizhu Powder is a classic ancient formula for treating spleen deficiency and diarrhea.It can strengthen the spleen,replenish qi,dissipate dampness and stop diarrhea,but its mechanism is not yet fully understood.This article takes Qiwei Baizhu Powder as the main research thread,exploring its ability to repair the intestinal barrier by regulating gastrointestinal hormones,digestive enzymes,tight junction proteins,and gut microbiota,in order to improve spleen deficiency and diarrhea.The aim is to provide theoretical basis and research direction for the application of Qiwei Baizhu Powder in animal production.In this experiment,two cycles were selected to prepare an animal model of spleen deficiency diarrhea.The model group mice were gavaged with senna leaf decoction(0.2m L/10g),and the appropriate model preparation cycle was selected by comparing the changes in body weight,D-xylose,and amylase of spleen deficiency diarrhea mice.KM mice were then randomly divided into 6groups: NC group,Model group,DSP group and Qiwei Baizhu Powder 5,10 and 20 g/kg groups.During the test,observe and record the weight,water consumption,food intake and active status of the test mice,use phloroglucinol method to determine the serum D-xylose level of the mice,use ELISA to determine the gastrointestinal hormone level and amylase content,use HE staining method to conduct pathological observation on the colonic mucosa tissue,and use immunohistochemical method and q PCR method to determine the expression level of Claudin-1 and Occludin protein,Using a combination of network pharmacology and bioinformatics methods to search for active ingredients and potential targets of action,and using 16 S r DNA amplicon sequencing technology to identify the gut microbiota.Results:(1)Compared with the blank control group,the 35 day model group showed weight loss,low activity,and decreased serum D-xylose and amylase levels,therefore,it indicates that the spleen deficiency diarrhea model has been successfully prepared.(2)The experiment found that Qiwei Baizhu Powder can significantly improve the mental state,activity,and water intake of mice with spleen deficiency diarrhea,alleviate pathological damage to their colon tissue,restore the structure of intestinal mucosal epithelial tissue,and upregulate the expression levels of Claudin-1 and Occludin proteins,increase the content of D-xylose and amylase in serum,and reduce gastrointestinal hormone levels.(3)149 active ingredients were selected from seven traditional Chinese medicines,including "Codonopsis pilosula,Poria cocos,Atractylodes macrocephala,Agastache rugosa,Muxiang,Glycyrrhiza uralensis,and Kudzu root",with a total of 367 targets.There are a total of 742 potential therapeutic targets for spleen deficiency diarrhea,with a total of 36 intersecting targets.Intersection targets mainly improve spleen deficiency diarrhea by regulating cell apoptosis,T cell receptor signaling pathways,inflammatory bowel disease,and TNF signaling pathways.(4)The diversity and abundance of gut microbiota in each group of mice treated with Qiwei Baizhu Powder increased.Conclusion: Qiwei Baizhu Powder can enhance the absorption function of the small intestine,regulate the secretion of gastrointestinal hormones and the content of digestive enzymes,regulate the intestinal microbiota,and protect the intestinal barrier from damage.It can play a role in treating spleen deficiency diarrhea through "multiple components,multiple targets,and multiple pathways".
