The growth of suckling piglets is an important part of the growth of commercial pigs.Weaning stress on piglets is often huge,it prone to a series of symptoms,including slow growth,high diarrhea rate and so on.However,using a wide range of antibiotics to ease stress is likely to cause drug residues in animals,bacteria resistance,and a serious threat to human health.Shen Ling Baizhu is a classic prescription of TCM spleen qi,nourishing qi to invigorate spleen,spleen dampness,qi stagnation,diffusing the lung and gas,Xingpi,check diarrhea and other effects,and has the advantages of no pollution and low residue.The experiment effect on piglet intestinal microflora and performance through the study of Shen Ling Baizhu,aims to explore the possible mechanism of Shen Ling Baizhu promote growth and alleviate the diarrhea of piglets,and provide reference for the application and promotion in clinical.This experiment selected 4 litters of newborn piglets from commercial farms and randomly divided into 4 groups,10 of each group(half male and female,the male and female in each group were randomly selected 3 piglets from each,and play a ear tag for fecal collection),The piglets and sows in the control group(K)were fed with conventional feed throughout the experimental period;in sow prevention group(M),the diets of the sows were supplemented with 0.3% Shen Ling Baizhu from the postpartum period to the end of lactation;in piglets long-term prevention group(ZC),the piglets had free access to Creep Feed with 0.3% SLB supplementation from birth to weaning;in piglets short-term prevention group,the piglets had free access to Creep Feed with 0.3% SLB 3 days before weaning.At 7:30~19:00 observe the defecation of piglets every day,counting the number of diarrhea piglets,in 3,6,9,14,21,28 and 35 days of age collected feces of the 6 marked piglets and extract the DNA by high-throughput sequencing,and eighing empty weight in the beginning and weaning of piglets.The result as follows:1.Alpha diversity analysis showed that piglet long-term prevention group had significant changes between short-term prevention group and control group at the 21 day old,and Chao1 and AC E index of samples of sow prevention group had significant difference with the control group at the 28 days of age(weaning age).And the C hao1 and AC E index were related to the age of suckling piglets.2.PCo A and NMDS analysis in the Beta diversity analysis showed that there were significant differences between the experimental group and the blank control group in the case of only considering the difference of sample evolutio n.The piglets intestinal microflora has the obvious difference in all groups in the case of considering both the differences of species evolution and division;Proving day age is a great influence factor of this difference again.3.Principal component analysis(PCA)showed that the effect of Sheng Ling Baizhu had significantly influence on piglet intestinal microflora4.Cluster analysis showed that the community structure of Firmicutes,Bacteroides and Proteobacteria in the intestinal tract of suckling piglets are dominant bacteria.The analysis to the level of the genus,Prevotella,Bacteroides which belong to Bacteroidetes and Lactobacillus which belong to Firmicutes asthe dominant genus.Thermography(Heatmap)and the change of dominant microflora analysis showed that Collinsella,Faecalibacterium,Butyrivibrio,Bifidobacterium and other intestinal benefit bacteria content distribution in the intervention group is higher than that of control group,but conditional pathogenic bacteria: Clostridium,Desulfovibrio,Odoribacter distribution is higher than the control group in each intervention group.5.Prediction of community functional genes showed that day age was also an important factor affecting the expression of functional genes.6.The rate of diarrhea and growth performance analysis showed that Shen Ling Baizhu has significant effect to reduce diarrhea rate of piglets and promote its growth.Conclusion:1.Shen Ling Baizhu can significantly reduce the diarrhea rate of piglets and promote the growth performance may be through improving the content of beneficial bacteria and decrease the content of conditional pathogenic bacteria to achieve.2.Day age is also an important factor to affec t piglet intestinal microflora diversity.
