The purpose of this paper was to alleviate the significant increase in feed cost caused by the rising corn price, this paper discusses the effect of wheat replacing corn and adding glucose oxidase on the production performance, reproductive performance, immune performance, antioxidant performance and intestinal physiology in Rugao yellow chickens before and after production. In order to provide a reference for the formulation of local laying hen diets before and after production. The first experiment was to explore the effect of wheat replacing corn on the growth performance, egg production performance, immune performance and intestinal physiology in Rugao yellow chickens before and after production. The experiment selected 27016-week-old Rugao yellow chickens with similar body weight and condition, randomly divided into 3 groups, each group had 6 replicates, and 15 chickens in each replicate were fed the following 3 kinds of diets:Corn group:Corn type(control group),50%Wheat group:50%wheat substituted corn type,100%Wheat group:100%wheat substituted corn type. The test results show that:(1)50%and 100%wheat replacement of corn both significantly increased the daily gain and average daily feed intake of 16~20 week-old Rugao yellow chickens, and significantly reduced the feed-to-weight ratio(P<0.05).There was no significant effect on the egg laying rate at 20-28 weeks and the number of eggs at 28 weeks(P>0.05)..(2)50%wheat replacement of corn and 100%wheat replacement of corn had no significant effect on organ weight, reproductive organs and reproductive hormone levels, and immune performance of 20-week-old Rugao yellow chickens(P>0.05).(3)100%wheat substituting maize significantly increased the ileum and cecum lengths of Rugao yellow chickens at the expected production date(P<0.05).Replacing corn with wheat had no significant effect on intestinal pH and intestinal enzyme activity(P>0.05).In conclusion, the replacement of corn with wheat can improve the growth performance of 16~20 week-old Rugao yellow chickens, but has no significant effect on the weight of internal organs, reproductive organs, immune performance and intestinal development of 20-week-old chickens. The second experiment was to investigate the effects of glucose oxidas on corn-soybean meal and wheat-soybean meal diets on the growth preformance, reproductive performance, antioxidant performance, immune performance and intestinal physiology of 16-20-week-old Rugao yellow chickens.A total of 360 16-week-old Rugao yellow chickens with similar body weight condition were selected and randomly divided into 4 groups with 6 replicates in each group and 15 chickens in each replicate.Corn group:corn type without enzyme,CG group:corn type+50 mg/kg glucose oxidase,Wheat group:wheat type without enzyme,WG group:wheat type+50 mg/kg glucose oxidase. The test results show that:(1)Feeding wheat-based diets significantly increased the average daily gain and average daily feed intake(P<0.05)of Rugao yellow chickens aged 16-20 weeks, and significantly decreased the feed-to-weight ratio(P<0.05).There was an interaction between glucose oxidase and the effect on average daily gain(P<0.05).(2)Lung weight was significantly increased by feeding wheat-based diets(P<0.05).(3)Glucose oxidase supplementation significantly increased the weight of small yellow follicles(P<0.05),and there was no interaction between dietary types and the effects of feeding glucose oxidase on reproductive organs(P>0.05);glucose oxidase supplementation could significantly increase plasma progesterone and estradiol levels(P<0.05),but there was no interaction between them on plasma reproductive hormones(P>0.05).(4)Feeding wheat-type diets significantly decreased liver total antioxidant capacity, catalase and glutathione peroxidase levels(P<0.05),and significantly increased malondialdehyde content(P<0.05).Glucose oxidase significantly increased liver total antioxidant capacity, catalase and glutathione peroxidase levels(P<0.05),and significantly decreased malondialdehyde content(P<0.05).There was an interaction relationship between the effects of diet type and glucose oxidase on hepatic glutathione peroxidase and total antioxidant capacity(P<0.05).Feeding wheat diet significantly decreased serum catalase, total antioxidant capacity, glutathione peroxidase and superoxide dismutase levels(P<0.05),and significantly increased malondialdehyde content(P<0.01);Adding glucose oxidase significantly increased the levels of serum catalase, glutathione peroxidase, total antioxidant capacity and superoxide dismutase(P<0.05),and significantly decreased the level of malondialdehyde(P<0.05).),but there was no interaction between the two on serum antioxidant indexes(P>0.05).(5)There was an interaction relationship between dietary type and glucose oxidase on the bursa index(P<0.05),but there was no interaction between the effects on the spleen index and serum immune index(P>0.05).The enzyme significantly increased the serum levels of immunoglobulin A,immunoglobulin G and immunoglobulin M in Rugao yellow chickens(P<0.05).(6)Feeding wheat-type diet and adding glucose oxidase significantly affects the length of the cecum(P<0.05),but had no significant effect on the length of the duodenum, jejunum, ileum, and rectum(P>0.05).Feeding wheat-type diet and supplementing glucoamylase had no significant effect on the intestinal pH value of each intestinal segment(P>0.05),and there was no interaction between the effects on the intestinal pH value of each intestinal segment(P>0.05).Feeding wheat-type diets and glucose oxidase had no significant effect on the levels of amylase and lipase in the jejunum and ileum(P>0.05),and there was no interaction between the effects on the levels of amylase and lipase(P>0.05).(7)Diet type and glucose oxidase supplementation changed the cecal microbial composition and relative abundance of Rugao yellow chickens.Feeding wheat diets significantly decreased the cecal microbial Shannon index,Simpson index and Chao1 index of Rugao yellow chickens(P<0.05).The ACE index was significantly decreased(P<0.05),and the addition of glucose oxidase had no significant effect on each index, and there was no interaction between the two(P>0.05).In summary, feeding wheat-based diets can improve the growth performance, reduce antioxidant performance, cecal microbial Alpha index and species richness of 16-20 week old Rugao yellow chickens. The addition of glucose oxidase can improve reproductive hormone levels, antioxidant performance, immune performance, cecal microbial composition and relative abundance of Rugao yellow chickens.
