Study On Tissue Culture Technology Of Xanthoceras Sorbifolia

Using seeds,stem segments and leaves as experimental materials,an efficient and stable tissue culture and propagation system was established through the optimization of sterilization conditions,the selection of different media and the combination and screening of the best exogenous hormones.The results are as follows:（1）the best way is to clean disinfection crown fruit seeds seed soaking natural cooling in the 70℃hot water,soak 4～5 h,with 5%of the surface of sodium hypochlorite disinfection first 8 min,then cut open kind of skin to take out the seeds,clean sterile water,75%alcohol immersion disinfection 30 s,clean sterile water three times,5%sodium hypochlorite disinfection 5 min,sterile water cleaning spare 5 times.The optimal medium for seed primary culture was MS+2.20mg·L^-16-BA+0.08mg·L^-1NAA+0.5mg·L^-1KT+30g·L^-1sucrose+6g·L^-1agar.The optimal medium for subculture was MS+0.5mg·L^-16-BA+0.5mg·L^-1IAA+30g·L^-1sucrose+6g·L^-1agar.The optimal medium for high growth culture of subculture was MS+0.5mg·L^-16-BA+0.5mg·L^-1NAA+30g·L^-1sucrose+6g·L^-1agar.（2）The most suitable disinfection method for stem segment is to sterilize with 75%alcohol for 30s and then 0.1%Hg for 9min.The optimal medium for stem segment induction was MS medium,and the inducing effect of hormones on axillary bud germination of stem segment was IAA>IBA>NAA.The optimal medium for axillary bud germination induction was MS+0.5mg·L^-16-BA+0.5mg·L^-1IAA,and the induction rate was 92.73%.MS+1.5mg·L^-1KT+0.5mg·L^-1NAA was the best medium for axillary bud proliferation and cluster bud induction.（3）The optimal medium for adventitious bud induction from leaves was MS+1.0mg·L^-16-BA+0.1mg·L^-1IBA,and the induction rate was 72.62%.The proliferation medium was MS+1.0mg·L^-16-BA+0.5mg·L^-1IBA+0.3mg·L^-1GA₃.（4）The optimal rooting medium for adventitious shoots was 1/2MS>WPM>MS,and the optimal rooting medium was 1/2MS+0.1mg·L^-1NAA+2.0mg·L^-1IBA,p H=5.85,dark culture changed to light culture for 5 days,relative humidity was 80%,the rooting rate was 83.56%,the average number of roots was 17.10,the average root length was 5.83cm,and the number of yellow leaves was the lowest.
